"Initiated as a part of the Bureau of Mines program for development of strategic minerals for national defense, various mines in the Poughkeepsie Gulch and Mineral Point mining districts were examined by C. R. Wilfley, W. R. Hamilton, .and N. E. Wvvlwy, Jr., Bureau Of 'Mines engineers, in July and August 1943, July 1944, and September 1945. In a subsequent report a proposal was made to evaluate this isolated area 'of some 15 square miles located in Ouray and San Juan Counties, Colo.Reconnaissance work in the area was begun in June 1946. Mapping and sampling were begun in July and was completed in Pougkeepsie Gulch and extended westward to include the Hendrick Gulch area of the east side of Ironton Park. Project work was recessed for the winter on October 15, 1946 after approximately 6 square miles of the Poughkeepsie Gulch,district been mapped and 801 samples had been taken.In this report the physical features, climate, and ore deposits districts are described, and a brief history of mining and production lead, zinc; and silver is given.The individual mines and prospects in, the area are described in detail.A list of patented claims and ownerships is given in appendix I, and analyses of samples are tabulated in appendix II.The San Juan preliminary project was directed by Paul T. Allsman, chief, Salt Lake City Branch, Mining Division and was supervised by William M. Traver, Jr., supervising engineer, Colorado Field. Office.Chemical analyses were made by the Salt Lake City laboratory under the direction of H.E. Peterson, chemist, and S. R. Zimmerley, chief, Salt Lake City Branch, Metallurgical Division.Harold A. "INTRODUCTION Direct visual microscopic determination of particle-size distribution and number concentration of atmospheric dust is time-consuming and trying to the eyes. By using the microscope to project images of the particles at high magnification onto a screen much of the eyestrain can be eliminated and particle-size distribution results obtained with less effort and at a considerable saving in time. It also permits simultaneous observation of the images by more than one person. Microprojection requires no more skill and very little more apparatus than is required in the use of the microscope. Anyone familiar with the microscope should be able to obtain as perfect images on the projection screen as in direct visual microscopy since the same laws and limitationsThe theory and practice of microprojection are described in detail in texts. 5/6/ The procedure has been used and found to be satisfactory for measuring the particle-size distribution of sub-sieve-size substances, such as pigments and powder 7/8/ in the same size range as that of atmospheric dust. The Bureau of Mines has used the microprojector and found it to be quite satisfactory for (1) the determination of the number concentration of atmospheric dust R.I. 3289 in samples collected by the Owens jet dust counter; (2) particle-size distribution in samples of atmospheric dust collected by the Owens jet dust counter and by the electric precipitator; and (3) the particle-size distribution of dusts, blood cells, bacteria, and pollen. Work is in progress at this Bureau on the use of the microprojector in counting impinger samples. The use of different kinds of illumination, such as reflected (dark field), polarized, monochromatic, and ultraviolet, in the projection of images of atmospheric dust particles is being considered."

The U.S. Bureau of Mines and others have conducted a number of occupational exposure surveys since the publication of the International Agency for Research on Cancer report of 1989(1), which labeled diesel exhaust as a probable human carcinogen. These surveys have assessed diesel exhaust exposure using measurement of diesel aerosol and/or specific polycyclic aromatic hydrocarbons as indicators of exposure. This report reviews, available diesel exhaust aerosol and gas measurement techniques. Measurement of human exposure to toxic agents is the first step in determining the exposure-dose-response relationship. It is a fundamental step in risk assessment. Measuring exposure to diesel exhaust pollutants is particularly challenging due to the complexity and variability of exhaust components. This paper reviews current occupational health standards for diesel emissions and available diesel exhaust gas and aerosol measurement techniques. Although surveys in mines have primarily assessed diesel exhaust exposure using diesel particulate matter (DPM) some exhaust gas measurements have been used as indicators of exposure. DPM is emphasized because it is a major component of diesel exhaust, is suspected to make a major contribution to the health hazard, and is measurable. OCCUPATIONAL HEALTH STANDARDS Occupational exposure surveys of diesel exhaust frequently measure regulated pollutants, or pollutants that are suspected carcinogens, even if no permissible exposure limits (PELs) have been established. In the United States, various organizations recommend a range of allowable values for diesel exhaust gaseous constituents, including carbon monoxide (CO), carbon dioxide (CO2), sulfur dioxide (SO.), nitrogen dioxide (NO2), nitric oxide (NO), and formaldehyde (HCHO). The United States Bureau of Mines has within its ranks a group of engi- neers who are engaged in working out quarry problems, the solution of which will be of general benefit to the industries involved. The problems undertaken are approached in a systematic and thorough manner, all the evidence for and against any proposed change or new development being collected as carefully as an attor- ney would prepare his data on an important case. Unlike the attorney, however, the Bureau engineer does not take sides; his judgment is based entirely and in- partially on the array of facts presented. In order to gain the desired infor- mation he visits many plants, consults with the leaders in the industry, works out experiments in his laboratory, and supplements this information with a stu- dy of all available literature already published on the subject in any part of the world. As an engineer who has for eight years conducted such inquiries in the stone industry, I propose to discuss the Bureau of Mines viewpoint; and it may be of interest to consider with me in a brief and general way the engineer's method of looking at quarry problems, and some of the results attained through past and present labors. While the Bureau has conducted no investigation directly for the crushed- st one industries, two years of investigative work at cement-plant quarries, and work now being conducted at lire-plant quarries, has a very direct bearing on the problems of the crushed-stone industry, since stripping, drilling, blasting, loading, and transportation are similar in many respects in all stone quarries. The results of cement-plant quarry investigations are to be found in Bureau of Mines Bulletin #160. Only limited work has been reported in the literature about the use of latex in soil. Unisol 91 2,3 (a 9:l oil-latex emulsion made b International Synthetic Rubber Company) and Phil1ips Petroset4 geotechnic emulsions are surface soil stabilizers which penetrate a maximum of two inches, but do not reduce permeability. Diamond Alkali 5 found that styrene-butadiene rubber latex as a one percent latex-soil mixture reduced permeability to 0.7 from 21 feet per year under a head of twenty feet of brine. We have found no literature reference for use of latex as sealant at depth in the soil, although asphalt has been applied at depth using civil engineering methods6,7. Most of the materials investigated as soil sealants in the present work were latexes, although some studies were also conducted with a variety of inorganic and organic chemicals. Of the latexes tested, most were rubber but a few were plastic, e.g. polyvinyl chloride. Sealants examined in this investigation are listed individually in Table I with supplier, code number, and other pertinent information. The materials consisted of the following general classes with additional details as indicated: 1.Anionic latexes: a. styrene-butadiene copolymers and terpolymers (SBR) b. butadiene-acrylonitrile copolymer (Paracrils®) c. polyvinyl chloride homopolymers and copolymers (WC) d. ethylene-propylene terpolymer e. vinyl acetate homopolymers and copolymers (VA) f. acrylics (acrylate copolymers) g. 2-chlorobutadiene homopolymer (chloroprene) (prepared specially as cationic material coagulated by soil). 2. 1. Introduction This document represents Part II of our final report on the development of a "Remote Sealing System for Extinguishing Coal Mine Fires. " It is a technical report which summarizes the results of system development and field demonstration. Part I of this two - part final report is an Operator's Manual for the application of the Remote Sealing System to a coal mine fire. 1.1 Background When coal mine fires go out of control and can no longer be fought directly, the standard fire fighting technique is to construct air-tight seals in the mine passages leading to the fire area so'--,that the oxygen supply to the fire is". cut off. Ultimately, it is desirable to isolate the fire zone in such a way that normal operations can resume in the remainder of the mine. The construction of seals in the- mine causes the atmosphere to pass through a toxic and explosive phase. Explosions, if they occur, are not only hazardous to the men working at the sealing operation, but can also undo much of the sealing work which has already been accomplished. Sealing of passageways surrounding the fire zone from within the mine is extremely difficult and dangerous. Methods for direct sealing are detailed in "Control of Mine Fires'. 1 An alternative to this approach is the technique of remote sealing. This involves drilling boreholes which intersect the mine passages at the locations where the desired seals are to be constructed. The seals are then constructed by pouring material down the borehole and filling a segment of the passage from bottom to top. During this operation, there are no personnel in the mine, all operations taking place on the ground surface.

The extrapolation of results from reconstructed laboratory soil columns to field conditions is difficult because of the limitations of laboratory experiments. Permeability and elution tests performed in the laboratory differed from the natural state in that the soil was sieved through 1/4 inch screen to remove large stones and roots, its density was controlled by tamping the soil into two-inch I.D. tubes instead of normal weathering, and water and latex were applied with a constant head of liquid. Tube wall effects are important in interpreting data. Conversely, interpretation of field results were confounded by non-homogeneities within the sample tested, a soil profile with gradation of permeability and density with depth, and uncontrolled weather conditions and seasonal changes that affected the results. Nevertheless, both laboratory and field tests showed that a specific latex did perform as a soil sealant. In view of these facts a soil box experiment was designed to overcome some of the disadvantages associated with the small laboratory soil columns and the field experiments. Two wood soil boxes 5 ft x 5 ft x 5 ft were built in the greenhouse at the Uniroyal Research Center. They were lined with plastic PVC sheeting provided with tubes every six inches of depth on one side of each box to measure water removal rates under saturated conditions. Tensiometer gauges were placed at depths of 6, 12, 18, 24, 36 and 48 inches and Bouyoucos blocs and resistance thermometers were positioned at 6, 24 and 48 inch depths. Each box was filled with a uniform sub-soil obtained from Ogdensburg, N. J. The texture of this soil was determined to be a sandy loam by the ASTM Hydrometer method, as shown below: [ ] The boxes were thoroughly wetted by sprinkling to abet settling and allowed to dry by draining to check seepage ports and functioning of tensiometers.

Inhaled particle contact with pulmonary surfactant in the hypophase lining pulmonary alveoli was modeled in vitro by exposing two respirable sized dusts, kaolin and silica quartz, to lecithin, a major component of pulmonary surfactant, emulsified in physiologic saline. Lecithin adsorbs to the dusts upon incubation at 37º C and suppresses their cytotoxicity as measured by pulmonary macrophage and erythrocyte assays, suggesting that pulmonary surfactant provides a defense system against prompt cell membrane lysis by inhaled dusts. Subsequent pulmonary macrophage phagocytosis and lysosomal enzyme digestion of the coated dusts was modeled in vitro by incubation with phospholipase A2 for one hour. With increasing lipase activity, silica hemolytic potential was restored to native silica levels. Kaolin was retoxified to levels far in excess of native kaolin hemolytic potential. Elution, thin layer chromatography and phosphate assay of treated dusts indicate most lecithin on silica is digested to lysolecithin and desorbed, silica retoxification being due to surface and adherent lysolecithin effects. Most lecithin on kaolin is not digested in this same time; that which is results in adherent lysolecithin which is responsible for the kaolin retoxification. Digestion with phospholipases A2 and C together produces only weakly retoxified kaolin. Results suggest that surface adsorption properties which control the adherence of prophylactic surfactant distinguish the pathogenic potentials of quartz, kaolin and mixed dust.

This contrasts with the strong potential for respirable quartz dust to induce pulmonary fibrosis and the much more limited potential of kaolin dust to do the same (MORGAN and SEATON, 1975).In an attempt to understand this anomalous situation, assays were performed on quartz and kaolin dusts incubated with dipalmitoyl lecithin emulsified in physiological saline, to simulate the initial contact of a respired dust with the pulmonary alveolar hypophase. The ability of kaolin dust to adsorb dipalmitoyl lecithin from physiological saline has been quantified (WALLACE, et al., 1975). Such lecithin treatment suppresses the cytotoxicity of both silica and kaolin to background levels (WALLACE, et a1.,1984).Surfactant coated detoxified dusts should then be phagocytised by pulmonary macrophages, incorporated into a macrophage secondary lysosome and exposed to hydrolytic lysosomal enzymes. Among these is phospholipase A2, which hydrolyses diacyl lecithin to lysolecithin. This was modelled by incubating lecithin treated dusts with phospholipase A2 in vitro to determine if the enzyme can digest the dust-adsorbed lecithin, and if the retoxification differs between the two dusts in a manner which might distinguish their pathogenicity."

To evaluate the potential for dust reduction, during blasthole drilling, of water or foam applied directly into the drill hole. Approach The Bureau of Mines tested a through-the-drill-steel water and foam injection technique in two noncoal mines to evaluate the method's potential for greater dust reduction. The tests were performed by drilling a series of blastholes for an entire face (a 90 minute time-period). For the first 45 minutes of drilling, foam or water was applied through the drill steel. Blowing ventilation was supplied to the entry about 50 feet from the face. The dust concentration measurements were made by a Real-Time Aerosol Monitor (RAM-I), and the data were read out on a strip chart recorder. The evaluation test results showed that the water or foam injection technique, when used on localized dust sources, can be an extremely effective dust control method. Small amounts of water used in this method have also allowed it flexibility to be used in water-soluble ores such as salt, gypsum, anhydrite, and trona. How The System Works Water or foam is mixed with compressed air (approximately 100-120 psi) with suitable metering systems and forced through the drill steel into the blast hole at the point of dust generation. Because the water or foam is applied at the source, minimal quantities of liquid are needed (0.2 to 0.6 gpm). These low flowrates are sufficient to just moisten the dust emerging from the blast hole, thus eliminating a major source of airborne dust. The wet drilling procedure also eliminates a second source of dust. With dry drilling, the dust from the blast hole falls to the floor. Since this can be a 15-ft drop or more, the impact can send the dust billowing up from the floor, adding to the airborne dust. With wet drilling, the moistened dust merely dribbles down the face.

"INTRODCICTIONSilicosis is the chronic fibrosing disease of the lungs caused by the inhalation of crystalline silica. However, inhalation of crystalline silica, may induce three distinctly different disease patterns; i.e., chrome silicosis, accelerated silicosis, and acute silicosis. These three disease patterns differ in their pathologic characteristics, clinical symptoms, onset of disease, and mortality. Many studies in the past have focused on the elucidation of mechanisms involved in the development of chrome silicosis. We recently reported that freshly fractured silica has surface properties that could make it more reactive with lung tissue than aged silica, and that this unique reactivity of freshly fractured silica may be involved in the pathogenesis of acute silicosis (Dalal et al., 1986; Vallyathan et al., 1988; Shi et II., 1988). Because freshly fractured silica could contain some moieties in an ""excited state,"" due to the breakage of silicon oxygen bonds, we postulated that the de-excitation of these moieties could produce light which could be monitored by a luminescence measurement technique. In addition, if this ""excited state'' of silica could trigger a greater biologic response by phagocytic cells, this activation could be monitored by a chemiluminescence technique in the presence of appropriate enhancers.This investigation describes for the first time the use of luminescence in the study of freshly fractured silica and its properties. Enhanced biologic reactivity of the freshly fractured silica with alveolar macrophages was monitored by the chemiluminescence technique.MATERIALS AND METHODSNative silica was obtained from the dust bank of the GenericRespirable Dust Technology Center, Pennsylvania State University, State College, PA, and ground for 3~ min in an agate ball mill. It was then sieved through a 20 micron mesh filter and used within 1 O min as fresh silica or stored in glass bottles for aging. X-ray energy spectrometry and X-ray powder diffraction studies on representative samples were made to confirm the mineralogic purity. All the silica samples were found to be 99 % pure with minimal detectable conlamination by metal ions."

"Respirable high-silica particles from four coal mines. a clay mine and a tunnelling site were analysed for silicon vs aluminum composition by performing scanning electron microscopy-energy dispersive X-ray analysis (SEM-EDS) at electron accelerating voltages of 20 kiloelectron volts (keV). The same particles were analysed at 5 keV to determine possible differences in surface vs bulk composition. In some cases high-silica particles exhibited a decrease in the ratio of silicon to aluminium K-alpha line intensities with decreasing electron beam accelerating voltage. This behaviour is consistent with aluminosilicate surface contamination or occlusion of a silica particle. Some 20 high-silica particles were analysed from each of 10 samples. Powdered aluminosilicate glass was used as a control for the behaviour of silica with a homogeneous distribution of aluminium in the particle. Significant differences were found between the control and sample means for an Illinois bituminous coal mine dust, two western and one central Pennsylvania bituminous coal mine dust low-temperature ashes. and a clay mine and mill dust sample, but not for an anthracite coal mine dust and two central Pennsylvania bituminous coal mine dust low-temperature ashes. INTRODUCTIONRESPIRABLE dust quartz content commonly is measured on bulk samples by using i.r. spectroscopy or X-ray diffraction. For research purposes, individual particle-by particle SEM-EDS analysis at 20-30 keV electron beam accelerating potential may be used also. Such measurements have not provided an unambiguous association of quartz content with occupational pulmonary disease prevalence in exposed workers (WALTON et al., 1977). Rather, there is evidence that coal rank modifies the relationship between dust exposure and pneumoconiosis development (WAL TON et al., 1977; ROBOCK and BAUER, 1990; ATTFIELD and MORRING, in press). SEM-EDS analysis was performed to measure the silicon fraction, Si/(Si +Ai), at 20 and at 5 keV on high-silica particles to determine if some respirable quartz particles generated by mining exhibit heterogenous composition with depth (WALLACE et al., 1990), which might affect the bioavailability of the quartz surface."

Considerable current interest is evident[l,2] in the use of transient electromagnetic methods for subsurface remote sensing. A transient method is discussed which uses a periodic video-type pulse containing a wide frequency range. A video pulse is used as an interrogating signal both for "target" identification and detection for relatively short depth (c 20 meters) applications in soil and soft rock media. If the medium is favorable, e.g., hard rock, greater depth applications appear to be feasible. The same procedures would be applicable for deep geological exploration but would require a substantial reduction in frequency and increase in target size. It should be noted that video pulse generators with 35 kv peak voltage and nanoseconds base widths are now within the state-of-the-art, thus significant power over broad spectral ranges is possible. In the design of a video pulse sounding system, 5 main problem areas can be identified; 1 ) selection of the pulse shape or spectral content of the video pulse, 2) design of a radiating structure which effectively couples such a pulse into the medium, 3) isolation of the radiating and receiving mechanisms, 4) realistically accounting for the dispersion and attentuation of the medium and the air-medium interface, and 5) interpretation or processing of the received signal waveform. Each of these problem areas will be discussed and illustrated with experimental and/or theoretical results. The Video Pulse Selection of the proper video pulse signal is dictated by 3 considerations, the maximum desired depth of penetration, the attenuation of the medium and the required resolution and size of the target. For a lossy medium, penetration and resolution are clearly at cross purposes and a compromise is necessary. The exact nature of the compromise is dictated by the application, but the spectrum of the video pulse should not contain any frequencies higher than the predicted cut-off, i.e., that frequency which is attenuated to the threshold level of the receiver. As a practical matter, it may be necessary to select 1 or 2 video pulses reasonably suited

Mine operators often rely on mine rescue teams to save lives during an underground emergency such as an underground fire, explosion or roof fall. It is extremely important that team members are provided with adequate exploration equipment and that training simulations are conducted in a realistic manner. A series of mine rescue training exercises was developed, conducted and evaluated by the National Institute for Occupational Safety and Health (NIOSH) in cooperation with the Pennsylvania Department of Environmental Protection, Bureau of Deep Mine Safety. The exercises were conducted at NIOSH’s Lake Lynn Laboratory during 1995 to 1998 and resulted in improved technology and training for mine rescue teams. For example, existing technologies were identified to help rescue teams during exploration. These included chemical light sticks to identify team members and mark crosscuts and mine materials, strobe lights for mapping out escape ways, and a helmet mounted thermal imaging system to see through smoke. A hands-free communication system showed potential for enhanced communications between team members and the fresh air base. A new high-visibility team lifeline that allows for flexibility of movement between team members was developed. A positive-pressure inflatable escape device (IED) was used to isolate the “hazardous” environment from fresh air and allow rescue team members to traverse through. An inflatable feed-tube partition that can rapidly block large openings such as underground passageways and simultaneously provide a feed-tube for high-expansion foam generators was also deployed for several simulations. During the simulations, mine rescue team members donned self-contained breathing apparatuses (SCBA) and traversed more than 305-m of mine passageways filled with nontoxic smoke. Visibility ranged from 0.3 to 0.9-m. In one area of the mine, a kerosene heater was used to simulate a fire and reduce the oxygen concentration. In another area, a 1.5 pct methane zone was established. The team members searched for "victims,” mapped the passageways, built temporary stoppings and ventilated the smoke-filled entries. The teams indicated that the simulations were challenging and very informative.

The United States National Institute for Occupational Safety and Health, through an informal partnership with industry, labor, and the United States Mine Safety and Health Administration, has developed and tested a new instrument known as the Personal Dust Monitor (PDM). The new dust monitor is an integral part of the cap lamp that coal miners normally carry to work and provides continuous information about the concentration of respirable coal mine dust within the breathing zone of that individual. Previous laboratory testing demonstrated that there is a 95% con?dence that greater than 95% of individual PDM measurements fall within ±25% of reference measurements. The work presented in this paper focuses on the relationship between the PDM and respirable dust concentrations currently measured by a coal mine dust personal sampler unit utilizing a 10 mm Dorr Oliver nylon cyclone. The United Kingdom Mining Research Establishment instrument, used as the basis for coal mine respirable dust standards, had been designed speci?cally to match the United Kingdom British Medical Research Council (BMRC) criterion. The personal sampler is used with a 1.38 multiplier to convert readings to the BMRC criterion. A strati?ed random sampling design incorporating a proportionate allocation strategy was used to select a sample of mechanized mining units representative of all US underground coal mines. A sample of 180 mechanized mining units was chosen, representing approximately 20% of the mechanized mining units in production at the time the sample was selected. A total of 129 valid PDM/personal sampler dust sample sets were obtained. A weighted linear regression analysis of this data base shows that, in comparison with the personal sampler, the PDM requires a mass equivalency conversion multiplier of 1.05 [95% C.I. (1.03, 1.08)] when the small intercept term is removed from the analysis. Removal of the intercept term results in a personal sampler equivalent concentration increase of 2.9% at a PDM measurement of 2.0 mg m-3.

1. Potential groundwater pollution resulting from large-scale surface mining in western states should be evaluated. 2. Criteria should be developed for the design, construction, operation, and abandonment of sediment ponds for maximum removal of suspended solids. 3. Methods should be developed for reducing suspended solids discharges from surface mines to less than 30 milligrams per liter. 4. A manual should be developed in the form of guidelines for sediment and erosion control planning and implementation. These guidelines must be designed to cover all phases of the coal surface mining operation (i.e., before, during and after mining). 5. A mine road manual should be developed for use by surface mine operators and State and Federal Agencies. Erosion, water control, and dust control should be covered in detail. 6. The block-cut, head-of-hollow fill, and mountain-top removal methods and techniques of contour mining should be evaluated, designed, and ultimately demonstrated in steep, mountainous terrain. Designs should be prepared for single and multiple seam applications. The demonstrations must show that these mining systems will secure rapid achievement of reclamation require¬ments and provide maximum, safe, economic coal recovery in an environmentally safe manner. 7. A Blasting Manual should be developed for use by industry and enforcement agencies. Stress should be placed on how to keep vibration levels and air blast pressures well below accepted safe criteria. Also, methods that can be used to control noise should be listed. 8. Investigations should be conducted on the effect of continued vibrations from blasting on structures that are near quarries. 9. Additional guidelines need to be developed for sampling and analysis of overburden material and associated coals for trace elements and noxious strata. Samples are extremely sensitive, and the source and manner of their collection, handling, storage, and treatment, as well as the techniques of analysis, need to be developed and described. This information can be used to predict possible future problems.

In a previous report, entitled A Gage and Recording Equipment for Measuring Dynamic Strain in Rock, a control circuit for a drum camera was described. The purpose of this control circuit was to provide a means for firing seismic caps, synchronizing the firing of the cap with the opening of the camera shutters and generating an electrical impulse at the instant of detonation. The operation of this circuit depended on the following two characteristics of seismic caps: (1) these caps contain a bridge, which breaks at the instant of detonation, and, the firing delay time of the cap is a function of the firing current. During the course of the Bureau's studies, considerable difficulty was experienced from "firing hash", i.e., spurious vibrations that appeared on the records at the time of detonation. The cause of most of this "firing hash" was traced to the intermittent grounding of the cap leads at the time of detonation. As the cap leads were still connected to the firing battery, the grounding of the leads permitted current to flow through the rock to the gage and to return to the firing battery through the shield on the gage cables. The intermittent flow of current along the cable shield produced potentials on the gage leads that were recorded as the "firing hash." To eliminate this difficulty, it is necessary to remove the firing voltage from the cap leads the instant the cap detonates. This report supplements the previous report and describes an improved circuit that can be used to fire seismic caps, remove the firing voltage from the cap leads the instant the cap fires, control the firing delay time in the cap, and generate reference signals at the time current starts flowing in the cap circuit and at the time the cap detonates. These reference signals can be used to determine the firing delay time of seismic caps and the time at which the seismic wave is generated by the explosion.

"INTRODUCTION A soluble mediator has been described which is released from activated rabbit leukocytes and induces histamine secretion from rabbit platelets (Barbaro and Zvaifler, 1966; Siraganian and Osler, 1971). This factor has been isolated, characterized and named platelet activating factor, PAF (Bcnveniste et. al., 1972). PAF has since been chemically identified as l-0-alkyl-2-acetyl-sn-glyceryl-3-phosphocholine (Benveniste et. al., 1979; Demopoulous et. al., 1979).PAF is a lipid mediator of broad biological activity. It causes smooth muscle contraction, contraction of lung tissue and decreased FEV 1; while enhancing capillary permeability thus causing edema (Pinkard, 1983). In addition, PAF is a chemoattractant and can lead to inflammation.PAF has been reported to stimulate the release of lysosomal enzymes, leukotriene B,, and lipid metabolites from neutrophils (Smith et. al., 1983; Shaw et. al., 1981; Lin et. al., 1982; O'Flaherty et. al., 1984). The objective of the present investigation was to further characterize PAP-induced stimulation of granulocytes and to develop a bioassay, i.e. chemiluminescence, for screening PAF receptor antagonists and inhibitors of PAP-induced activation of granulocytes. The possible use of the chemiluminescence assay to monitor PAF synthesis by human granulocytes is also described.METHODSBlood was drawn from healthy volunteers into a blood storage bag containing CPD anticoagulant. Granulocytes were partially purified by dextran sedimentation and further purified by centrifugal elutriation as described previously (Jones et. al., 1980). Granulocytes obtained by this procedure were approximately 95 percent pure. The number of granulocytes in this preparation was determined using an electronic cell counter (Coulter Model Z1, Coulter Instrument Co., Hialeah, FL). Isolated granulocytes were used to monitor membrane potential, hydrogen peroxide and superoxide release, oxygen consumption, calcium uptake and, in some cases, chemiluminescence. Studies of PAF receptor antagonists were conducted using neutrophils isolated by density centrifugation. Briefly, blood was layered onto Mono-Poly-Resolving Medium (Flow Laboratories, McLean, VA) and centrifuged at 22 °C and 300 g for 30 min. Neutrophils collected from the second band of this gradient were at least 90 percent pure."
