Metalliferous sediments and/or saline brines have now been discovered in numerous sea floor deeps along 900 km of the axial rift zone of the Red Sea (Fig. 1). The Atlantis II Deep remains the most interesting of the the brine deeps both in terms of intensity of submarine hydrothermal processes and in terms of potential economic importance. In fact, successful completion of a pre-pilot mining test in 1979 indicates that the Atlantis II Deep may become the first deep sea base metal sulfide mine (Mustafa and Amann, 1980). Under the most optimistic scenario, the Atlantis II Mine might be established 4-6 years after completion of a year long pilot mining test in 1983-84. If established, mining would produce about 100,000 tons per day of wet mud with about 200 tons per day contained zinc, and proportional amounts of Cu and Ag. Expected mine life would be about 15-20 years (Anon., 1981). In addition to direct economic importance, the Red Sea brines and metalliferous deposits provide significant insight into exploration for sea floor brine-related ore deposits and for analogous ancient deposits. In this paper I will concentrate on the economic potential of Red Sea deposits and sea floor exploration for brine related deposits, but some ramifications of relevance to ancient deposits are extant.

In January of 1969 the United States Department of Health, Education and Welfare, through the National Air Pollution Control Administration, published a series of Air Quality Criteria (4'S) and Control Techniques (6,7) for air pollutants in compliance with the 1967 Clean Air Act which will affect the operation of all fuel burning equipment in the very near future. The design of equipment for the future which will use fossil fuels will also be affected. The purpose of this paper is to discuss some of the modifications which might be anticipated in the future for fuel burning equipment. One class of the several air pollutants looked at first by the USDHEW was the sulfur oxides. Other pollutants, such as nitrogen oxides, will be studied and controls for these will supposedly be written in due time. The obvious use of low-sulfur fuels, coal, oil and natural gas is suggested by the Control Technique(6). Conversion to other sources of power, such as nuclear energy and hydropower, is also suggested. The statement is made that since possible hydroelectric sources are limited and nuclear plants are not yet available on a large scale, the changeover to these sources of power will not be overnight. The bulletin (6) suggests that one possible solution in some areas may be the use of low sulfur coals for short periods of time during periods of severe air pollution. A curve showing the effect of this was given in the 1969 April issue of the Power magazine, where A. E. Boyer and F. B. Kaylor of Stone and Webster also described a computer program called DOSAGE which might be used to check weather, temperature, humidity, wind direction and velocity, and sound an alarm when that 0.1% of the time per season is here that requires the use of low-sulfur fuel. The evolution of open-pit mining has been rapid during the last two decades. The development of larger, more productive and efficient equipment has been one of the key factors in this open-pit advancement. Selection of the proper equipment and its correct usage is paramount to successful operation while maximizing return on investment. The quantity of material being moved prior to mining the saleable product is increasing at a significant rate. For example, at a southwestern copper mine more than two hundred million tons of over- burden have been removed prior to exposing an ore which contains less than 1% copper. A recent survey of twenty-three Western Hemisphere copper mines reveals the average strippingratio was 2.2:l with a range of 0.9-8.5:l. In the face of increased mineral demand, the resulting technological developments and rising overburden to ore ratios, many open-pit mines now use totally different concepts for the removal of ore and overburden. An increasing number of these concepts now being installed include wheel tractor scrapers as a major part of the material handling system for unconsolidated overburden removal. To say scrapers move only unconsolidated overburden is incorrect. On the contrary, ripped phosphate rock, copper ore, iron ore, and moderately consolidated overburden are also being moved by scrapers in various areas of the world. As reported in the Minerals Year- book, however, about one-half of the material moved in metal and non-metal mines is never drilled and blasted or broken by other means. We will, therefore, in this paper, concentrate on the role of wheel tractor scrapers in unconsolidated overburden; the area of greatest usage.

Although the markets for coal in the United States have changed rapidly over the last three decades, the uses of coal, namely, to produce heat and power or coke have been left almost unchanged. The coke is used either as a chemical reducing agent alone or as a reducing agent and source of heat. Most of the heat and power markets for coal require no coal processing, although for more than 50 years, many proposals to skim the more valuable substances in coal before burning have been advanced. Thus, the only large tonnage market requiring processing of coal is high-temperature carbonization to make coke. At present, coal requires processing for some small markets, but for future potential large markets, such as gasification, conversion to liquid and gaseous fuels, and conversion to chemicals, coals will require more processing than is now being done.

Today man is taking his family back to a cave saying, "Come enter in because of what I have done". At the close of World War II Germany said if they had gone underground one year earlier with more of their operations, the war could have lasted four years longer with perhaps much of Britain devastated. The protection of the home front no longer rests with the soldier in actual combat. The battle will be won at home with the most careful kind of protection against missiles. After Nagasaki-Hiroshima Sir Winston Churchill said in substance "Up to now the Almighty has mercifully withheld from man the knowledge of these terrible weapons of destruction."

CONTAC Engineers is a Chilean based company which works on process control and automation, principally for the minning industry. Jointly with other local and international engineering firms, CONTAC has participated in most of the last expansion and modernization plans being carried out at the Chilean pinning industry. At presente, CONTAC is involved on a research and development plan for the design and implementation of new instrument and process control procedures for mineral process automation. One of these efforts is the design of an ore size analyzer for coarse ore being transported over a conveyor belt. A feasibility and prototype study was made at a SAG Mill Plant and currently a high resolution, high speed analyzer is being developed. It will became available on early 1986.

Resource Enterprises, Inc. (REI) is exploiting the coalbed gas resource by working in unison with active underground mining operations. At REI?s pilot project at the Soldier Canyon Mine, Carbon County, Utah, REI has drilled nearly 15,250 m of horizontal in- seam and slant boreholes during the initial two years of drilling activity. Produced gas has been sold to a natural gas transmission company on a continuous basis since mid 1983.

To determine the effect of flux crystal size on the production rate and strength of sinter, batch sintering tests were conducted on samples of both dolomite and limestone of various crystal sizes. In addition, calcination tests .were performed on the dolomite and Limestone to determine the effect of crystal size on calcination rate at various temperatures normally attained in sinter beds. The results indicate that flux crystal size does not affect sinter production rate or sinter strength.

The Devonian salt basin, which contains the Saskatchewan Potash deposits, extends from just east of the foothills in Alberta, north as far as the Peace River area, across Saskatchewan and into Manitoba as far east as Range 10 west of the First Meridian and south into Montana and North Dakota. The basin is closed everywhere except to the northwest. The known potash deposits are confined almost entirely to the Province of Saskatchewan, with the exception of a small area in western Manitoba bordering the Saskatchewan boundary. The following discussion will concern only the Saskatchewan part of the basin. The evaporite series in the basin is defined as the Prairie Evaporite Formation of the Elk Point Group, of Middle Devonian age. Recent work done by potassium-argon dating methods has indicated an Upper Middle Devonian (Givetian) age of from 265 to 347 million years for the potash. The Elk Point Group consists in ascending order of the Ashern, Winnipegosis and Prairie Evaporite Formations. The Ashern formation, sometimes called the Third Red Bed, consists of dolomitic shales or shaly dolomites. The Winnipegosis is a reef type dolomite, usually with good porosity, and in many cases oil-staining, although to date no production has been obtained. The Prairie Evaporite formation consists of anhydrite overlain by halite and finally, in Saskatchewan, by the potash beds. In almost all wells which have been cored for potash the lowest potash bed is underlain by one or two bands of anhydrtte, which are used as marker horizons. Cores from a few wells which cored to the bottom of the salt series have shown flecks of potash throughout the halite.

The listed title of my talk to-day is "MODERN ON STRAND SINTER COOLING". Perhaps it should he ve been "COMPLETION OF SINTERING ON THE STRAND" as the two are synonymous. By the very nature of the DWIGHT-LLOYD SINTERING PROCESS partial cooling on the strand is always present, but not completion of sintering and cooling on the strand. I have mentioned the Dwight-Lloyd process just to be certain the type of sintering under discussion is fully identified because some of the basic mechanisms of sintering occur in various pyro-processing of fines lying in close quiescent contact, such as powdered metals practices, some types of pellet induration, firing of briquets, or other compacts, where the entire bed is brought to, and held at, the high reaction temperatures of sintering. Figure 1, The continuous sintering process as originally conceived consisted of intimately mixing carefully proportioned burdens, placing them in a comparatively thin, draft permeable layer, on a moving grate and igniting the surface while drawing sir through the bedded burden. The moving grate was advanced slowly as the line of burning fuel travelled from the surface to the bottom of the bed and no more fire zone could be seen.

"As mine operators continuously look for ways to improve functionality and increase capacity of their facilities, existing process equipment is often pushed to and past its designed limitations. As a result, equipment manufacturers are asked for economical solutions to meet redefined processing needs. Newmont USA (Newmont) in Nevada was faced with such a need in their Carbon-in-Leach (CIL) train. In a collaborative effort between Newmont and Derrick Corporation, tests were conceived to assess the viability of drop in replacement screen using a three dimensional screen panel. The urethane pyramid screen panel was designed to increase flux while maintaining the existing equipment footprint. The test is the end result of an ongoing research and development project that Derrick undertook in an effort to refine and upgrade their urethane screening media. The application of this new screening media is presented for this CIL application along with future plans to implement the technology in trash, safety, carbon dewatering, carbon sizing, and other scalping applications.INTRODUCTION In Spring of 2014, Process Machinery Associates (PMA), Derrick Corporation’s (Derrick) local area representative, met with Newmont USA (Newmont) to discuss the operation of the in tank screens in the carbon in leach (CIL) train at Twin Creeks, near Golconda, Nevada. The process was originally designed for 13,000 +tons per day (tpd) but has been steadily rising since the plant was commissioned and now Newmont requires 16,000 tpd to be processed. The original screens were running at their maximum capacity and Newmont required additional screening capacity to continue the upward trend in production. Physical separation by use of screening is one of mineral processing oldest methods. Screening media can be prone to blinding and pegging and the necessary support structures required for finer apertures result in smaller open areas. Therefore, more screening area generally requires larger panels and therefore larger machines, screening media that doesn’t blind, or a combination of both. In an effort to avoid larger machines, Derrick Corporation had been working on a revolutionary three dimensional panel which would add non blinding screen area in the same footprint of an existing machine. Alpha testing of the new screen panels began at Newmont Mill 2/5 (Carlin, Nevada) in July of 2013 and was still occurring when PMA approached Twin Creeks about beta testing a refined machine and pyramid panel. After presenting the initial results, Twin Creeks determined that an upgraded machine and pyramid panels would be a possible solution to meet production needs. The testing of a machine eliminated the risk of capital costs to Newmont and provided Derrick the opportunity to obtain additional data."

SCHEME DESIGN London’s Heathrow Airport, as the world’s busiest international airport, is missing one vital aspect - a dedicated rail link which connects directly to the city. All this is about to change with the construction of Heathrow Express - a fast, high quality rail service between the airport and the centre of London. It was recognised in the mid 1980s that if Heathrow was to accommodate the forecast growth in passenger numbers and the resulting increase in road traffic, a dedicated airport rail service would be vital. The main access into the centre of Heathrow is via a road tunnel with a direct link to the M4 motorway. The airport has introduced a number of traffic management schemes, but these can only achieve so much benefit. Growth in air passenger traffic is predicted to continue at an average of 4% per annum well into the next century and it was clear that significant investment was needed. Heathrow Express at a cost of over £350 million will provide this when it starts its service in June 1998. The business case for the scheme is based on a rider ship of around 15% of air passenger traffic. The initial Heathrow Express service will be based on four trains per hour, up to 8 car train lengths, providing a high speed non-stop service between the airport and central London in just 16 minutes. Initially a joint venture between the British Railways Board and Heathrow Airport, by mutual consent of the two parties, Heathrow Airport recently purchased the British Railways Board share making this one of the largest private sector rail projects currently under construction in the UK. Leaving Paddington, the main rail terminus in West London, Heathrow Express trains will travel 12 miles on the newly electrified line to airport junction, north of the airport. At this point they will leave the mainline to enter a 5 mile tunnel section that runs underneath the heart of the airport. The railway companies are responsible for track work, signalling and electrification of the 12 mile main line section including a grade separated junction over the main tracks for the Great Western Railway which links London and South Wales. Procedures for particle size analysis on respirable dust from coal mines have been evaluated. Using respirable dusts dispersed in a laboratory dust chamber, size distributions measured using cascade impactors have been com- pared with the results of a variety of laboratory sizing methods performed on samples collected on filters placed side-by-side with the impactors. The laboratory techniques included centrifugal sedimentation, laser diffraction/scattering and automatic particle counting. The general agreement among the various methods was very good, with systematic differences which can be attributed to the different definitions of size used. Simple conversion factors have been obtained which can be used to transform the laboratory data to equivalent aerodynamic sizes. Similar comparisons have been made on samples collected in underground coal mines. Again, the results are in quite good agreement although small discrepancies were observed in some cases. These have been attributed to some degree of agglomeration in the airborne dust, leading to slightly coarser apparent distributions in the impactor samples.

Asbestos is a generic term for industrially useful fibers recovered from exploitable deposits of the asbestiform varieties of five silicate minerals: chrysotile, anthophyl-lite, actinolite-tremolite, cummingtorute-grunerite, and rie-beekite. Asbestos and asbestiform fibers of some other minerals are known to be carcinogenic. Minerals crystallized in the asbestiform habit-are characterized by unique properties (morphology, strength and flexibility, physical and chemical durability, and defect-free surface structures). The key to the unusual properties, and health effects, of asbestiform fibers appears to be in their unusual surface structure. In contrast, non-asbestiform crystals of the same minerals, including the five so called asbestos minerals, do not have these properties and are not carcinogenic.

Introduction The Gold Quarry gold deposit is the largest bulk tonnage epithermal gold deposit in northeastern Nevada to be developed by Newmont Gold Co., formerly known as Carlin Gold Mining Co. Exploration and development of the deposit extended over a period of nearly 50 years before bulk tonnage mining became economically feasible. Proven reserves at Gold Quarry are currently estimated at more than 255 t (8.2 million oz.) of gold, accounting for more than 40% of the total Carlin Trend mineable reserves for 1986. Milling began in late 1985; dump leaching of lower grade ores began in May of 1986. The mill has exceeded the initial design throughput rate of 6.3 kt/d (7000 stpd) and is now processing more than 8 kt/d (9000 stpd). The dump leach facility will process more than 7.3 Mt (8.1 million st) of lower grade ores in 1986. The Gold Quarry ore body is to be developed by an open pit that will eventually be 1830 m (6000 ft) long, 1220 m (4000 ft) wide and 300 m (1000 ft) deep. The mine currently produces 77 kt/d (85,000 stpd) on a 24-hour-a-day, seven-day workweek. Planned pit production will increase to nearly 168 kt/d (185,000 stpd) in 1988. Production equipment currently includes one Demag H-/285, three Komatsu PC-1500 hydraulic shovels, two Cat 992C loaders and 21 90 to 135 t (100 to 150 st) haul trucks. A prototype, self-contained mine refuge station air supply sys- tem was tested in an underground metal mine to verify it's ability to keep oxygen and catbon dioxide levels at acceptable concentrations in an emergency situation. In this test, 25 volunteers were assembled and sealed in a functional underground refuge station for a period of 24 hours, without the benefit of the standard com- pressed air supply. The prototype system was designed to: 1) operate on a self-contained power supply for a period of at least 27 hours; 2) supply oxygen at a metered rate; and 3) remove car- bon dioxide by re-circulating refuge station air through soda lime scrubbing beds. During the test period, the catbon dioxide concentration remained at an average of 2500ppm and oxygen concentration stayed between 20.2% and 20.6%. The volunteers found the prototype easy to use and seemed confident in its ability to provide a safe atmosphere. The test showed that this technology could become part of an underground mine's refuge station emergency equipment.

Slickensides cause potentially hazardous ground conditions in underground coal mines. Investigations by MSHA’s Roof Control Division indicate that many slickensides represent bedding plane faults and drag folds that formed during coal bed decollement, a style of faulting that occurs in shale units above coal seams. These slickensides formed in response to regional tectonic compressional stresses, rather than by differential compaction, and are parallel to folds and thrust faults. This paper relates slickensides in the Appalachian, Illinois, and Black Warrior basins to the structural grain of the Appalachian Mountains, provides descriptions of slickensides, and advocates geologic mapping as a predictive method for identifying areas with adverse ground conditions.

Brought on stream in 1975, the 10,000-tpd copper oxide processing plant of Anamax Mining Co. at Twin Buttes, Ariz., is now at design capacity. Based on acid agitation leaching, solvent ion exchange, and electrowinning, the Anamax plant bridges an ore to metal gap by producing mine site copper cathodes from lime-bearing oxidized feed. Until recently, such material was considered submarginal because copper recovery was uneconomic using conventional methods. The new Twin Buttes oxide plant, the largest of its kind in North America, is one stage in the expansion of Anamax, a joint venture owned equally by The Anaconda Co. and Amax Inc. Other phases include an accelerated stripping program in the pit, an expansion of the sulfide concentrator from 32,000 to 40,000 tpd, and construction of a 57 MW diesel generating station for the oxide plant. Once the basic oxide flowsheet had been selected, Anamax awarded a contract to a joint venture of Arthur G. McKee and Co.' (Western Knapp Engineering Div.) and Davy Powergas' (a member of the Davy International group). The contract covered process design, engi¬neering, procurement, and construction. The property, identified by Banner Mining Co. in the 1950s and developed by Anaconda, was put into production in 1965 as an open pit mine equipped with a 32,000-tpd sulfide flotation concentrator. The Twin Buttes mine has since become one of the largest earth¬moving operations in the world, excavating some 10 tons of waste per ton of ore. At the original sulfide mill feed rate, the mining fleet moved 375,000 tpd of material (225,000 tpd alluvium and 150,000 tpd of rock). The most significant copper mineral in the ore body is chalcopyrite, which is scattered in pockets of weak mineralization. Cap oxidation has progressed to depths of several hundred feet, principally as chryso¬colla. Present estimates place minable reserves at 347 million tons of sulfide ore grading 0.6% copper and 55 million tons of oxide ore grading 0.9% recoverable copper from 1.25% total copper. The nonre¬coverable fraction is composed of native copper and insoluble copper oxides and sulfides. The oxide ore has been partially stripped, and 25 million tons having a copper content greater than 0.6% have been stockpiled. The stripping of oxide ore to expose sulfides will continue for several years. The copper oxide studies launched by Banner Mining in the 1950s were continued by Anaconda. Process routes investigated included: acid leaching; cyanide leaching; segregation roasting; chlorination, oxidation, and reduction; flotation in combination with leaching; cal¬cite flotation plus leaching; copper oxide flotation; and sulfidization flotation. By late 1968 test work favored a low temperature roast followed by an ammonia-ammonium carbonate leach of the ore. During this period, acid leaching had been discounted because of excessive con¬sumption of expensive acid by the high limestone ore matrix. Restric¬tive SO2 emission controls in 1971 suggested an increase in acid production and availability at a lower cost. Research then switched to acid leaching, resulting in development of the present flowsheet. Mechanically agitated leaching in a cascaded series of tanks was selected in preference to vat leaching after piloting a stirred-tank system. Preliminary testing of vat leaching indicated that inter-vat solution clarification problems could be caused by gas evolution from leaching of high limestone ore. Lack of research time for resolution of this problem, plus success of the pilot agitation system, dictated the final flowsheet selection. In 1973 Anamax decided to proceed with agitation acid leaching to produce liquor for a solvent ion exchange (SIX) and electrowinning (EW) installation. At that time, such systems were proving their eco¬nomic viability, and they allowed direct recovery of high quality cath odes while bypassing the smelting and refining of lower grade cement copper precipitates. The Anamax oxide plant processes a grade of about 1% recovera¬ble copper from stockpiled sources and new mine production. Unit operations include size reduction, agitation leaching, countercurrent decantation washing of residue, pH adjustment of solution, clarifica¬tion, solvent ion exchange, and electrowinning of copper, with a mini¬mum of environmental impact. The feed to the primary crushers is composed of: 1) -10-in. stockpiled oxide ore, which has passed through the mine crusher and has been grade-segregated to allow blending for a consistent feed material. 2) -4-ft ROM oxide ore stockpiled in the early mining operation. 3) -10-in. ore currently being mined. Ore is delivered in 100-ton mine trucks to a dump pocket, where an apron feeder transfers the ore to an inclined 6-in. fixed grizzly. The grizzly oversize passes through a 48 x 60-in. primary jaw crusher, where it is reduced to -6 in. The fixed grizzly undersize and the crusher product are conveyed to a 4-in. vibrating grizzly. Oversize from this grizzly reports to a 7-ft Standard cone crusher, where it is reduced to -1 in. The crusher product and the vibrating grizzly undersize are conveyed to two 8 x 20-ft double-deck vibrating screens that operate in closed circuit with two 7-ft short head cone crushers. Screen undersize (-1/2 in.) is delivered to a covered conical fine ore stockpile with a designed live capacity of 15,000 st. Beneath the stockpile are ten Pioche belt feeders feeding two conveyor belts that supply the milling circuits. Each group of five feeders has two variable speed and three fixed speed drives. Grinding to 95% -48 mesh is done in two parallel lines, each consisting of an 111/1 x 18S -ft rod mill and a 12' x 30-ft ball mill in open circuit. Water is added to maintain a pulp density of 72% solids in the mills. Further water additions are made to the ball mill discharge sumps to create a 60% solids slurry for transfer to the leaching reactors. The nominal grinding rate is 440 tph solids. A splitter box on the product from the mills separates the incoming slurry into two streams, one for the leaching reactors and the other for the pH adjustment reactors. This split is determined by the lime content of the feed material and the pH of solution entering the pH adjustment reactors. Five 30-ft diam by 31-ft high mechanically agitated, rubber-lined leach tanks are arranged in cascade, with gravity transfer from one to another by enclosed launders. Concentrated sulfuric acid (93.2%) from storage is added to the pulp principally at the first leach tank, and the reacted slurry leaving the cascade after a 5-hr residence time at 50% solids is pumped to countercurrent decantation (CCD) wash thickeners.' At this point, most of the soluble copper has been leached from the ore. Up to 250 lb of sulfuric acid is consumed per ton of ore, the acid being supplied in tank cars from a nearby smelter. Leached slurry at 50% solids is washed countercurrently with return raffinate from the SIX plant in a series of four Doff-Oliver center caisson, 400-ft diam thickeners. Solids advance from thickeners No. I through No. 4 to the tailings dam. SIX raffinate advances from thickener No. 4 to thickener No. l, where a pregnant solution overflows to pH adjustment. To optimize solvent extraction, the pH of the pregnant solution from CCD thickener No. I is adjusted from 1.5 to 2.5 by adding unreacted ground slurry (split from the grinding circuit) to the preg¬nant overflow of No. I CCD thickener. The resulting slurry is retained for 45 min in three mechanically agitated leach tanks. INTRODUCTION Proliferation and enhanced synthesis of collagen by pulmonary fibroblasts have been shown to be key steps in the development of chronic silicosis (Goldstein and Fine, 1986). The regulation of lung fibroblast proliferation by cytokines released from alveolar macrophages may be an important pathogenetic mechanism in the development of the fibrotic process (Kelley, 1990). One cytokine, platelet-derived growth factor (PDGF), promotes fibroblast proliferation by inducing the movement of quiescent (Go) cells into the C1 phase of the cell cycle (Chen and Rabinovitch, 1989). Others regulate the rate of transition of fibroblasts from Gl into the S phase (Leof et al., 1982). These two classes of cytokines have been termed, respectively, competence and progression factors. One approach used to examine the release of cytokines from macrophages is the fibroblast proliferation assay in which fibroblasts are exposed to culture supernatants from macrophages exposed to various stimuli. In most of these assays, the supernatant contains fetal calf serum which provides the competence factor(s) necessary to facilitate the proliferation of fibroblasts (Bitterman et al., 1982; Bitterman et al., 1983; Elias et al., 1988). Recently, a fibroblast proliferation assay using plateletpoor plasma (lacking competence factor(s)) as a substitute for fetal calf serum has been described (Kuman et al., 1988; Bauman et al., 1990). In this assay, the release of a competence-inducing PDGF-like growth factor from rat and human macrophages can be distinguished from other cytokines that act as progression factors. In order to obtain more consistent results and with the ultimate goal to be able to discriminate between the effects of competence factors as opposed to progression factors, we have conducted experiments to determine the appropriate concentrations of plasma and PDGF required for imparting competence in the fibroblast proliferation assay. We tested lung fibroblast cells obtained from explants of rat lung tissue and also a fetal human lung fibroblast cell line obtained from American Tissue Culture Collection (ATCC153). MATERIALS AND METHODS Fibroblasts Specific pathogen-free, male Sprague-Dawley rats were use in some studies. Animals were given a lethal intraperitoneal dose of sodium pentobarbital. Fibroblasts were isolated by chopping the lung in enzymes that digest the connective tissue but liberate lung cells for further study (Rabovsky et al., 1989). After digestion, the remaining lung tissue suspension was filtered through two layers of sterile gauze and centrifuged to recover lung fibroblasts. These were resuspended in culture medium that contained 10% fetal calf serum and distributed to culture plates for growth. In other experiments, a human fetal lung fibroblast cell line, obtained from American Type Culture Collection, Rockville, MD, 20852, was used instead of rat lung fibroblasts. In these cases, a 1 ml ampule containing human fetal fibroblasts was plated into a tissue culture flask containing medium plus 10% fetal calf serum. For both types of fibroblasts, culture medium was changed 3 times per week and cultures were incubated at 37°C until confluent. Harvested rat and human lung fibroblasts were quantified using an electronic cell counter equipped with a cell sizing attachment (Coulter Electronics, Inc., Hialeah, Florida). Tritiated Thymidine Incorporation The basic procedural outline of Kumar et al. (1988) was used with modifications to evaluate tritiated thymidine incorporation into fibroblast DNA following exposure to PDGF and plasma. Both rat and human lung fibroblasts were plated at 50,000 cells/ml at a density of 250,000 cells/25cm2 culture plate. Cells were quiesced for 4 days with 2% rat plasma. As the assay was refined, fibroblasts were quiesced in plasma-free media for 48 hrs, since the mitogenic activity of 2% plasma was variable. Test medium was applied for a period of 6 hrs, followed by a 24 hr tritiated thymidine (lµCi/ml) labelling period in plasma-free media. Medium alone was used as a negative control and media with 10 or 20% fetal calf serum was used as the positive control for rat and human fibroblasts, respectively. Cell Quantification and Measurement of Mitogenesis Twenty-four hours after the addition of tritiated thymidine, the fibroblasts were washed with 5ml of fresh serum-free media, centrifuged and resuspended in phosphate-buffered saline. The cells were dissolved in 0.5m1 of O.1N NaOH and radioactivity determined in a beta counter. Incorporation of trititaed thymidine as an index of DNA synthesis was expressed as DPM/fibroblast.
