Mineral resource studies by the Bureau of Mines indicate three areas within the Selway Bitterroot Wilderness have mineral resource potential. Regional studies suggest that a granitic stock on the south side of the Wilderness has high potential for a Climax-type stockwork molybdenum deposit (pl. 1, no. 8); but detailed surface investigations failed to recognize a deposit. The Elk Summit area on the north side of the Wilderness contains subeconomic resources of columbium (niobium)-bearing ilmenite in placers (p1. 1, no. 19). Subeconomic silver-copper-lead resources occur in a vein on the northeast side of the Wilderness at the Clift Mine (pl. 1, no. 2-5). The study area has no known deposits of oil and Haas, coal, geothermal resources, or other energy-related commodities.

"Continuous thickening by an operation called sedimentation is the separation of suspended solid particles from a liquid stream by gravity settling. The primary purpose of thickening is to increase the solids concentration of the feed stream. High rate thickeners are designed specifically to maximise the flocculation efficiency. A detailed investigation has been carried out to understand the influence of ore type and operating parameters such as bed level, bed mass, rake arm torque and flocculent concentration on thickening operation. Tata steel’s Noamundi plant data shows that variation in ore type, and operating parameters are major contributors to fluctuation in thickener performance. A lot of research has been done earlier on thickening operation but effect of ore type and operating variables by using plant data was never touched before.In this context, the performance of thickener was evaluated by daily data to study the influence of ore type and operating variables on thickener operation. Investigation shows that friable ore % in feed mix is proportional to slime density because friable ore is porous & brittle in nature. As a result, friable ore generates more fines/ultrafine during communition process in wash plant. Increase in flocculant concentration and bed mass is showing a positive change on high rate thickener performance Investigation shows that optimum bed mass and flocculant concentration is required to achieve desired pulp density in thickener underflow at different feed pulp density (g/cc).INTRODUCTIONOver the years, Iron Ore Beneficiation at Tata Steel, Noamundi has seen varied changes. Depletion of high grade iron and with increasing iron ore throughputs coupled with lowering of the hard ore a subsequent increase in slime generation, slime management and clear water requirement for wet Plant has thrown up myriad challenges. To overcome these challenges, many improvements initiatives were taken by Tata Steel in last decade. One of the major changes is the recent High Rate Thickener commissioning at Tata Steel Noamundi to cater the high iron ore throughput requirement. Slime reporting to Thickener has increased from 1.4 MTPA 2.5 MTPA over the last few decades. The above changes further aggravate the poor settling and dewatering characteristics of the slurry which is being feed to HRT. Considering the above mentioned constraints and stringent environmental implication proper water management has become a major challenge to reckon with. The thickener is used to increase the concentration of the suspension by sedimentation, accompanied by the formation of a clear liquid. In most cases the concentration of the suspension is high and hindered settling takes place."

LLOYDMINSTER is the centre of a well developed agricultural area, lying on the Alberta-Saskatchewan interprovincial boundary in Township Fifty. The surrounding country is gently undulating, without any notable topographic features, but twenty-two miles to the north the North Saskatchewan river, and thirty miles to the south the Battle river, have cut deep valleys. In the spring of 1934, a wild-cat well (Lloydminster Gas Company's No. 1 well) was successful in striking a large flow of gas at a depth o 1,974 ? feet. The gas was piped into the town, a short distance south, and Lloyd-minster became the first Saskatchewan town to be served with natural gas. Following this discovery, other wells were sunk and, up to the present, thirteen wells have been drilled m what is here designated the Lloydminster area. Ten of these wells are in Saskatchewan and three in Alberta. Seven were successful in obtaining gas, three oil, and three have been unsuccessful. The area tested by this drilling is about eight thousand acres, six thousand of which can be considered as proved gas or oil land. To the south and west of Lloydminster, in the Province of Alberta, there are several areas in which oil and gas have been discovered. The Wainwright field lies nearly fifty miles to the southwest; Battleview, where recently gas has been discovered, is thirty miles west-southwest; and the Ribstone area is about thirty miles south and a little west. At Ribstone, a number of wells were drilled some years ago and there is a small production of oil. The data relating to the wells in the Lloydminster area that are recorded in this paper are taken from the records of the several companies operating in the field. The names of these companies are given on the map and on the type stratigraphic section (Figures 1 and 2). The author wishes to record his appreciation to Mr. R. H. Shaw and Mr. 0. C. Yates, both of Lloyd-minster, for much valuable information; also to Mr. E. Swain, of the Department of Natural Resources, Regina, with whom the writer has visited the field on several occasions, and who, by his enthusiasm and encouragement, has done much to forward the development of the area. The well samples were examined at Ottawa by officers of the Borings Division of the Geological Survey of Canada, who also made the interpretations of the logs. This paper addresses a gap in longwall multi-seam interactions and their predictions, providing a case study for future multi-seam operations for both anticipated conditions and a method for understanding the changes in stress regime. Multiple panels were modelled with the results analysed and compared to panel experiences to validate the parameters and properties. The software chosen (LaModel) is a numerical modelling package designed to model stresses and displacements on thin deposits. It utilises a displacement-discontinuity variation of the boundary-element method and has been used extensively throughout both the United States and Australia in multi-seam operations. Analysis of the results demonstrates a clear correlation with the above workings and regions of significant strata instability. This provides the opportunity going forward to analyse new models and incorporate into mine planning and geotechnical designs. Continuous site-based assessment should be undertaken at multi-seam operations to better understand and quantify likely vertical stress levels within the workings. With a detailed analysis of models prior to extraction and a review of models post extraction. Further research is required as LaModel is limited to vertical stress and it is likely that interactions impact both the horizontal and shear stress regime. 3-dimensional modelling could further assist in this understanding, combined with a thorough stress monitoring regime. "The Pierre Beauchemin gold deposit, situated 15 km NW of Rouyn-Noranda, produces an average of 1050 tonnes of ore a day. Reserves are 1.2 Mt grading 5.6 g/t Au. The deposit, located in a shear zone within the Flavrianpluton, in the Blake River Group, is hosted by four major rock types: meta-trondhjemite, meta-tonalite, hybrid rocks and meta-diorite (the prefu; ""meta-"" is omitted thereafter). The trondhjemite, tonalite, and metamorphic hybrid rocks are spatially distributed as a series of sub-horizontal tabular sills that form a genetically related group. Early fractures that permitted either the emplacement of pre-diorite felsic dikes or the formation of a visible alteration caused by early fluid circulation might have been developed in response to initial strain created during regional deformation. These fractures were precursors to the emplacement of diorite and the development of major shear planes. Sub-horizontal north-trending en échelon diorite lenses are observed at the mine. Together, these diorite lenses constitute larger scale diorite zones which are observed at the scale of the pluton. The diorite zones are interpreted as evidence of an episode of sub-vertical extension sustained by the layered trondhjemite, tonalite and hybrid system. The anisotropy created by the less competent sub-horizontal en échelon diorite was an important factor. In shear zone development and subsequent mineralization. The tonalite and diorite accommodated further defomation and developed the main N030°/40° ESE shears along with the spatially associated N060°/50°-60° SSE, and the N000°/10°-40° E shear zones. The N000° shears are interpreted as refracted or deflected equivalents of the N030° shears. The combined shears compose the Eldrich shear system which trends approximately N030°/40° ESE and has had a general reverse movement with a minor sinistral component.Gold mineralization occurs in all four major rock types as en échelon sigmoidal shaped lenses associated with the major N030° deformation system. The distribution of ore lenses can be divided into two categories: The first category occupies sub-horizontal diorite zones. The lenses trend N-S, dip 100 40° E, and are associated with shear zones, sub-horizontal fractures or tonalite-diorite contact planes. The second category of lenses is found in felsic granitic rocks; the lenses trend either N0300!40°ESE or N060°/50°-60° SSE. Local hydraulic breccia zones are observed near ore zones occupying felsic granitic rocks. These breccias may have resulted from high fluid pressures brought upon by confinement as a result of impermeable zones in an anisotropic tonalite-diorite system."

Deep Vibro Techniques namely Vibro Compaction (VC) and Vibro Replacement (VR) are widely used in ground improvement to densify sands and strengthen cohesive soils. This paper describes the application of these techniques for the construction of 20 Ha Iron Ore Stockpile Yard with the maximum stockpile height of 14 m in Quang Ngai, Vietnam. The challenges of this project are to limit the stockpile allowable settlement and the surrounding pile’s lateral movement. The soil condition can be summarized as 12m – 18m thick of loose to medium dense SAND with qc around 5-18MPa and followed by 2.0 – 6.0 m thick soft clay with qc around 0.4 – 0.8MPa. Localized very dense sand layer was found. VR using stone columns were installed in soft clay layer while VC was adopted to densify sand layer. Additional shallow stone columns were built in sand strata to minimize pile top lateral movement. The post construction performance was verified by a large 6 m by 12 m plate load test (PLT). The predicted PLT deformation using Priebe’s improved properties for the VR will be presented and compared with the measured plate load test results.

Optimized mixer settler designs will now handle total flows of over 6,000 m3/hr.Computational Fluid Dynamics (CFD) is a timely and predictable tool for design development. Lightnin has the capability of using numerical analysis methodology to evaluate flow patterns and phase separation. CFD can show performance differences between changes, in mix tanks, in settler geometry, and in liquid depth, against variable flow rates. Understanding the differences allows the development of optimized designs. This paper focuses on the major design components of the mixer settler and how technological advances will lead to further improvements, when designing for today’s largest flow plants.

Hydrothermal sulfides were recovered from the16°50?S triple junction area in the North Fiji Basin, at a water depth of ca. 1900 m. The chimney samples can be divided into three groups according to their major metal contents: 1) type 1 (Fe-Cu-rich), 2) type 2 (Fe-Cu-rich with minor Zn), and 3) type 3 (Zn-rich) chimneys. Type 1 chimneys are mainly composed of chalcopyrite and pyrite, and are enriched in elements commonly precipitated under high temperature conditions (> 300°C), such as Cu, Co, Mo, and Se. Type 3 chimneys consist dominantly of sphalerite and marcasite with traces of pyrite and chalcopyrite, and are enriched in elements commonly associated with low temperature (150 to 250°C), such as Zn, Cd, Pb, As, and Ga. Type 2 chimneys show mineralogy similar to that of type 1 chimneys but their metal contents range between type 1 and type 3 samples. Trace element compositions of basaltic rocks indicate that magma generation in the triple junction area was influenced by two different sources: N-MORB and E-MORB. Sulfur and lead isotope patterns of the hydrothermal chimneys show distinct differences between the type 1 and type 3 chimneys. For example, type 1 sulfides are depleted in 34S and have lower Pb isotope ratios compared to type 2 and 3 chimneys. The d34S values (0.4 to 5.6?) of chimney sulfides are typical range of sulfur isotope composition of sediment free mid-ocean ridge hydrothermal system. Type 2 and 3 sulfides show slightly higher d34S values than type 1 sulfides, which can be explained by subsurface boiling of hydrothermal fluids followed by mixing of saline solutions with ambient seawater. Lead isotope compositions of the chimneys indicate that the Pb in type 1 and type 3 chimneys originates from hydrothermal leaching of N-MORB and E-MORB volcanic rocks, respectively. Considering that both these chimneys are located in the same area, our model suggests that type 1 chimneys formed by hydrothermal circulation controlled by an intrusion of N-MORB magma; the fluids were not influenced by boiling during the formation of type 1 chimneys. In contrast, type 3 chimneys are younger and were formed by hydrothermal circulation closely related to E-MORB magma intrusion at shallower depth. The necessary conditions for phase separation of the hydrothermal fluids that formed the type 3 chimneys might result from changed P-T conditions in a shallower high-temperature reaction zone beneath the hydrothermal venting site. Under these conditions the fluid may cross the two-phase boundary for seawater, ensuring subcritical phase separation. The mineralogical and geochemical properties of these chimneys suggest that type 1 and type 3 chimneys were probably black smoker and white smoker, respectively. Type 2 chimneys might be product of low-temperature replacement of type 1 chimneys, which is related to formation of type 3 chimneys.

Structural Controls Interpretation of regional aeromagnetics, radiometrics and satellite image data has identified three structural domains (fig 2): The Fairyland Domain: a general N - S structural trend. The Cracow Domain: a general NW - SE structural trend. The Bald Mountain Domain: a general N- S structural trend. Known mineralisation is confined to the Cracow Domain in which there occurs two major NW - SE trending fault zones. The Cracow Fault Zone (CFZ) and the Immy Creek Fault Zone (ICFZ). Enhanced detail of the CFZ (fig 3) shows three main structural trends within it: NW-SE structures which include the mineralised White Hope South fault zone and the Golden Mile fault zone. NE-SW structures which have an influence on mineralisation in the Orange Creek, Fernyside and Golden Plateau areas. N-S structures comprising several major structural zones.

"A new method for MOCVD has been developed in which a precursor solution is directly injected into a reaction chamber. The vaporization of short pulses of liquid is facilitated by atomization with a piezoelectrically-operated nozzle. The system has been used successfully to grow oriented, thin Ti02 (rutile) films on sapphire substrates with the crystalline relationship (101)[010]R I (1120)[0001]s. A growth rate of approximately 2 monolayers per pulse was achieved at 650°C for a reactant impingement rate of about 800 monolayers per pulse. Also, epitaxial LiTaO3 thin films were grown on (0001) sapphire from a single source, bimetallic precursor, lithium tantalum hexa-t-butoxide as the precursor. X-ray diffraction shows that the LiTaOa c-axis is normal to the sapphire surface. The x-ray pole figure and crosssection TEM diffraction show that the films were also in-plane oriented, with sapphire [1010] parallel to lithium tantalate [1010]. These films are shown to have extremely low optical attenuation, and refractive index that compares well with bulk values.IntroductionMetalorganic chemical vapor deposition (MOCVD) techniques have come into increasing use in recent years, due to the growing versatility and availability of precursors and improved environmental advantages over the halogenated compounds used for conventional CVD. Most commonly, the films are deposited by dissolving the metalorganic reactant (typically a metal alkoxide) in a carrier gas, which is carried through heated lines to a heated substratel . The metal alkoxides decompose by pyrolysis, leaving the films on substrates. While this method has been used to produce excellent epitaxial and heterepitaxial films, it has the disadvantages of complexity: the precursor must be heated to fix its vapor pressure, the gas flow rate carefully controlled, and the carrier lines heated to prevent condensation before reaching the reactor. Additionally, this conventional method cannot be used with certain low-volatility precursors that decompose upon heating."

THE illustration below shows the design of the face of the clock to be erected as a memorial to the American engineers who gave their lives overseas in the World War. It will be placed in the tower of the library at Louvain, which has been rebuilt through contributions from America. Large clocks seldom have figures on their faces, and each hour on this one will be marked by a gold star, the four faces containing 48 stars, one for each State of the Union. Contributions toward the memorial have been received from every one of the States, and from Alaska and Hawaii as well, but additional subscriptions are invited by the Com-mittee which is arranging for the memorial.

Chlorination of organic matter by direct injection of chlorine gas or hypochlorite has been investigated metallurgically worldwide with little understanding of the basic chemistry of the passivation mechanisms in- volved. In this paper, chlorination of carbon is attributed to oxidation-reduc- tions, resulting in surface halogenation of graphitic or amorphous carbon. It is proposed that chlorine and carbon interact to form thin coatings of chioro-carbon complexes that block the adsorption of gold cyanide com- plexes, thus passivating the gold adsorption sites of carbonaceous matter. Analytical, spectrographic, microscopic and electron-optical methods have been applied to demonstrate the proposed mechanism of chlorine-carbon bonding, as well as to evaluate the advantages and limitations of chlorination in the pretreatment of refractory carbonaceous gold ores.

The Blast Furnace Studies Section, Metallurgieal Division, Bureau of Mines, has been working for several years on the mechanism of desulphurization of iron and steel by slags. The usual assumption that desulphurization takes place by diffusion of FeS from metal into slag and subsequent reaction of this FeS with CaO to produce CaS has seemed to us inconsistent with a number of experimental results we have obtained. We undertook, for example, to determine the proportion of sulphur in a slag present as FeS. An entirely satisfactory method of doing this has not yet been evolved. The results of extraction with metallic antimony led to an interesting conclusion, however. FeS is readily soluble in metallic antimony, either physically or by chemical reaction. On the other hand, CaS was found to be insoluble. Hence it would appear that by rapid extraction of the slag with antimony we might expect to determine the distribution of sulphur between iron and calcium. The results showed that much of the sulphur was present as FeS. In some instances the extraction of sulphur as FeS was almost quantitative. For example, in a slag containing 3.8 grams Fe and 3.7 grams S, extraction removed 1.18 grams S and 2.12 grams Fe. If all the iron present in the antimony had been FeS, 1.21 grams S would have been extracted. In another slag with 8.6 grams Fe and 0.06 gram S present, 0.14 gram Fe and 0.08 gram S were extracted. This was obviously extraction of sulphur as FeS within the analytical error and showed that FeS was present either in solution or as a dispersion. Attempts to determine the solubility of FeS and CaS in slag by quenching and ultramicroscopic investigation have indicated that the solubility of FeS is very low, and that reactions in the slag are probably not to be considered as homogeneous. A more detailed report on these theoretical considerations will be made at a later date.

El presente trabajo señala la geología, litología y mineralización del yacimiento minero El Tapado del distrito Yanacocha, situado al norte de Cajamarca. Este se encuentra conformado por diferentes depósitos auríferos epitermales de alta sulfuración, los cuales se desarrollan dentro de un vulcanismo calco-alcalino de composición andesítica a dacítica de edad Mioceno y son correlacionables con la Formación Porculla del Grupo Calipuy. La geología del depósito está conformada por rocas porfiríticas (intrusiones) y flujos piroclásticos tales como tufos de cristales, tufos líticos y brechas piroclásticas afectados por eventos eruptivos de brechas freáticas - freatomagmáticas. Todas estas rocas han sufrido posterior alteración hidrotermal con ensambles típicos de sistemas tipo ácido-sulfato. La mineralización económica está controlada por procesos hidrotermales y se encuentra asociada a sílice masiva y sílice vuggy ( 1-10 g/t Au y estructuras de hasta 136 g/t Au ) y en menor grado sílice alunita y sílice granular (moderada a baja ley).

Several works have been dedicated to study the ballistic response of multilayered armor systems (MAS), including those using natural fiber composites as second layer. These composites disclosed good ballistic performance, having economic and environmental advantages. In the present work, the objective was to investigate the effect of areal densities of each MAS layer, in the overall performance of a MAS with an alumina ceramic front, a curaua fabric reinforced composite filling, and an aluminum alloy backing the target. Ballistic “backface signature” tests were conducted, varying the areal densities of the MAS layers according to a Box-Behnken design. The results showed a similar influence of the ceramic and aluminum layers in the MAS “backface signatures”, although deformation and failure mechanisms were significantly different. The composite also showed significant participation in the overall MAS performance. Since the composite is cheaper and lighter, a balance minimizing the other layers would be advantageous.

A vehicle proximity warning–collision avoidance system is being developed using GPS and wireless local area networks to improve safety of off-highway trucks in open pit mines. After two-and a-half years of research, software development, and laboratory testing, field tests were carried out at operating limestone quarries and open pit mining operations to evaluate GPS accuracy and overall system effectiveness under rugged operating conditions. This article describes the progress made on this NIOSH-funded project and discusses the GPS accuracy tests and overall system tests that were carried out during the year 2001 at the Colorado School of Mines Survey Field and at the Morenci open pit copper mine in Arizona. The design of hydrostatic liner for a deep potash mine shaft in Saskatchewan poses tremendous technological challenges. Technology has evolved to a point where one could consider a new innovative approach to hydrostatic liner design. But there is still a place for conventional technologies such as cast iron tubbings. As will be discussed in this paper, improvement to manufacturing process, installation procedures and even new materials can bring this technology to an excellent level of performance. With the spur of new projects in the potash industry of Saskatchewan, the authors worked in various shaft liner designs from the conceptual stage through to the construction phase. They were either directly leading the design work on some of these projects or acting as an external reviewer on others. Most of these shaft liner designs involved cast iron tubbing technology and ground freezing. The major challenge in sinking potash shafts in Saskatchewan is to go through a water-bearing ground formation under high pressure, known as the Blairmore or Manville fm. The only proven stabilizing technology for these formations is ground freezing. For a potash shaft, dry shaft conditions are highly desired, so a fully waterproof hydrostatic liner is required. All the existing potash mine shafts in Saskatchewan, except for two with composite steelconcrete lining, used the cast iron tubbing technology. Two new shaft tubbing liners have been constructed recently, one in the Athabasca Basin and one for a potash mine in Saskatchewan. When the authors began working on these new projects, what became quickly evident is that the technological knowhow for designing and constructing tubbing liners was no longer available within Canada and that the past construction methods used to sink tubbing lined shafts was obsolete. The tubbing liner for the shafts sunk in the sixties were designed either by German or British companies. In both of these countries only few tubbing lined shafts were sunk in last 40 years and the technology used in the sixties was not improved. There are only few countries where tubbing is routinely produced and installed: China, Russia, Ukraine and Poland. Out of these countries Poland is the one where this technology is the most advanced. The authors, working in close collaboration with engineers from Poland, attempted to rebuild this knowledge for the mining industry in Canada. This process involved thorough analysis of old technical reports, literature review and study of current polish design and construction practices as well as site visits of existing shafts. Through actual construction experience, some lessons were learned as well as ideas for improvements. In this paper, the authors will present a succinct review of design and procurement process. But, the practical aspects of the construction and installation methodology will be treated with more emphasis, more specifically the practice leading to good liner performance. Some of the poorly understood technical aspects of tubbing installation will be discussed more thoroughly and some currently accepted myths dispelled.

Investigations of world wide coal face strata control practice, the study of theoretical calculations and detailed engineering analysis have been combined to design and select a powered roof support suitable for use in conjunction with the extraction of 4.5-m (14.7- ft) of the 7.5-m (24.6-ft) thick Warwickshire Thick Coal Seam. The interface between geotechnical criteria and engineering design has been closely considered along with the following 3 important factors: Firstly, the need to further improve strata control and operational characteristics whilst producing a wall integrated overall face design. Secondly, to enable the supports to be rapidly transported from face to face in roadways driven at the minimum size for operational requirements. Finally, to provide hydraulic power, automatic control and monitoring systems which will improve operations reduce manpower requirements and improve management information. Clear design requirements were determined in order to allow selection on a commercial basis. A method of tendering is described with the subsequent analysis and selection process. In the event oversupply of coal in the UK resulted in the order not being placed. Brief consideration has been given to supports suitable for reserves to be mined In a few year's time where there is potential for extraction well in excess of 4.5-m (1 4.74.

"INTRODUCTIONOrganics extractable from respirable diesel exhaust soot particles by organic solvents have been known for some time to be direct acting frameshift mutagens in the Ames Salmonella typhimurium histidine reversion assay (Huisingh, et al). Upon deposition in a pulmonary alveolus or respiratory bronchiole, respirable diesel soot particles will contact first the hypophase which is coated by and laden with surfactants. To model interactions of soot and pulmonary surfactant, we dispersed soots in vitro in the primary phospholipid pulmonary surfactant dipalmitoyl glycerophosphorylcholine (lecithin) (DPL) in physiological saline. We have shown that diesel soots dispersed in lecithin surfactant can express mutagenic activity, in the Ames assay system using S. typhimurium TA98, comparable to that expressed by equal amounts of soot extracted by dichloromethane/ dimethylsulfoxide (DCM/DMSO)(Wallace, et al, 1987). Here we report additional data on the same system using additional exhaust soots and also using two other phospholipids, dipalmitoyl glycerophosphoryl ethanolamine (DPPE), and dipalmitoyl phosphatidic acid (DPPA), with different ionic character hydrophilic moieties. A preliminary study of the surfactant dispersed soot in an eucaryotic cell test system also is reported.MATERIALS AND METHODSSoots D4 and D6 were collected at two different times as scrapings from the exhaust pipe of a diesel tractor-trailer immediately after the engine was stopped. Surfactant dispersions were made by ultrasonic probe sonicating 25 mg of commercially obtained DPL, DPPE, or DPPA into 1O ml of O.85% physiological sterile saline (PSS) for 10 min at 40 watts power. For surfactant dispersions, 2.5 mg soot was mixed into 2.5 ml of the appropriate phospholipid dispersion. Mixtures were sonicated by 40 watts sonicator probe for one min, then incubated in the sonicator water bath for 30 min at room temperature; the mixture was then incubated for one hour at 37C while being rotated continuously. Dilution series were made with saline. For solvent extraction, 2.5 mg soot was mixed into 2.5 ml of DCM and incubated in a sonicator water bath for 30 min at room temperature. The sample was then mixed with 2.7 ml DMSO, placed in a dry bath at 45C, and the DCM evaporated under a stream of nitrogen gas until 2.5 ml of liquid remained. Ames salmonella/microsome assay utilizing TA98 for frameshift mutation was used for mutagenicity testing, with and without S9 activation, as described by Ames et al (1975). Unfiltered sample extractions or dispersions were preincubated with tester cells for 90 min at 37C : 0.1 ml of sample was mixed with 0.5 ml of PSS, or of S9 mixture, and 0.1 ml of an overnight TA98 culture incubated samples were then added to 2 ml of top agar supplemented with biotin and a trace amount of histidine, and overlayed onto Vogel-Bonner minimal media plates. After 48 hours incubation at 37C, revertant colonies were counted. Solvent controls were DMSO, DPL, DPPE, DPPA, and saline. Positive controls were trinitrofluorenone (1 ug/plate), for samples not treated with S9, and 2-aminoanthracene for S9 treated samples (2.5 ug/plate). All samples were assayed in duplicate. Sister chromatid exchange (SCE) assay was performed, following Lett, et al (1981), using V79 cells, of Chinese hamster pulmonary fibroblast origin. Stock diesel sample suspensions were made as 10 mg soot/ml DPL suspension, or 20 mg soot/ml in DMSO. Aliquots of these soot preparations were added to 15 ml of culture medium per sample flask to result in concentrations of 25, 50, and 75 ug soot/ml medium. After 12 hours incubation, 200 ul of 25 uM 5-bromodeoxyuridine (BrdU) was added to each flask. After another 35 hours incubation, 200 ul of 10 ug/ml colcemid were added; and the cells were harvested three hours later. Cells were swollen by hypotonic saline, and fixed twice with methanol/acetic acid, and burst on microscope slides. Cells on the slide were stained with Hoechst 33258, exp"
