"Summary: In this report the differential regulation of NF-KB and nitric oxide (NO) was investigated in the mouse macrophage cell line RAW 264.7 following exposure to a mineral dust (silica) and /or an endotoxin (Lipopolysaccharide, LPS). The results indicated that silica and LPS can significantly induce the activation of NF-KB as well as elicit enhanced production of NO in RAW 264.7 cells as part of an early inflammatory response mechanism. A 24-hour time-course study showed that NO release from these cells continued to increase following the initial stimulus by LPS or silica. In contrast, activation of NF-KB was maximal at 6 hours and then showed a steady decline to 24 hours. The production of NO was suppressed by protease inhibitor and antioxidant, both of which block the activation of NF-KB. Surprisingly, the use of an NO synthase inhibitor resulted in an enhancement of NF-JCB activation. These findings suggest that NO produced in macrophage cells in response to an inflammatory stimulus like silica or LPS my be linked to a negative feedback role on the activation of NF-KB. It is now accepted that nitric oxide (NO) plays a pivotal role in the inflammation response process when macrophage cells are activated by a pro-inflammatory stimulus (l). Production of NO is dependent on the activity of the enzyme nitric oxide synthase (NOS). Up until now, at least three distinct genes have been described which encode different isoforms of NOS. NOS has been cloned from a variety of different mammalian cells, including constitutive neuron NOS, constitutive endothelium NOS and inducible NOS (2). Macrophages appear to be the principal cellular source for the inducible form of NOS (iNOS) activated in response to an inflammatory producing event. The mechanisms subserving the regulation of the iNOS gene and NO production have been under intense investigation in recent years (3-5). The Bureau of Mines investigated the feasibility of utilizing low-grade phosphatic shale as a feed for phosphoric acid production. Present practice in the phosphate mining operations of southeastern Idaho is to remove and either stockpile or waste low-grade phosphatic shales as the high-grade phosphate beds are selectively mined. Acid digestion studies were performed on uncalcined shale and on shale calcined to remove free carbon to determine phosphoric acid quality, yield, and amenability to filtration. The untreated shale, containing 21.4 pct P205, was digested with sulfuric acid in a single-tank reactor to yield phosporic acid and gypsum residue. Although 96 pct P205 recovery was attained in digesting uncalcined shale, only the calcined product gave satisfactory filtration rates. Calcined shale yielded phosphoric acid recovery of about 93 pct of the contained P205. It was higher in impurities than commercially prepared acid derived from high-grade western phosphate rock. An acid filtration rate of 745 lb P205/ft2/day was attained. This value is lower than the rate of 900 lb P205/ft that is generally considered to be acceptable in industrial production of phosphoric acid from high-grade phosphate rock.

In August 1943 the Bureau of Mines completed seven churn-drill holes in an area of old mine workings near Linden, Wis., formerly known as the Argall mine. The work was undertaken as part of a general investigation by the Bureau of the zinc and lead producing area of southwestern Wisconsin at a time when special effort was being made to increase the production of metals necessary for war. The area was selected for investigation on the basis of its favorable geologic structure, past history, and apparent mineralization.

Fiber optic systems are being deployed in locations where explosive gas atmospheres are normally present or are present under fault conditions. The National Institute for Occupational Safety and Health, Pittsburgh Research Laboratory (NIOSH, PRL) conducted a study of laser safety in potentially flammable environments. Researchers conducted experiments to estimate the mean and standard deviation of laser powers needed to ignite 6% methane-air atmospheres using single mode optical fiber tips covered by two types of iron oxide (Fe304 and (FeMn)203) mixed with a ceramic adhesive. The iron oxides, heated by a 1064 nm continuous wave laser, ignited the methane-air mixtures ,at similar powers. The minimum igniting power and maximum non-igniting power (10 tests) were 407 and 350 mW, respectively, using a 62.5 pm fiber. Laser beams guided by 125 and 80 pm diameter cladding single mode fibers produced similar methane-air igniting powers. Ignition was not observed using coal particles at powers that produced ignition with the iron oxides. Threshold ignition delays using the single mode fiber were approxinlately proportional to the inverse square of the igniting power. Ignition delays were significantly longer than the reported activation time for a comnlercial fiber optic power limiter. Comparisons are made with the results of other researchers. The Bureau of Mines is currently conducting a comprehensive study to evaluate all aspects of recovering gold from large, low-grade deposits in the Golden Sunlight area, Whitehall mining district, Jefferson County, Mont. Prior to World War II this area produced significant quantities of gold. The study includes geologic mapping, diamond drilling, assaying, computer analysis of assay data, metallurgical research and ore testing, mine and mill planning, and an economic evaluation of all factors bearing on the establishment of a producing gold mine. Preliminary evaluation of the deposit indicates about 31 million tons of submarginal gold resources. American Exploration and Mining Co. (AMEX) has assisted the Bureau in this project by providing drill site preparation, control assays, and geological aid for the Bureau's exploration work and by furnishing comprehensive assay information of the property and copies of base and geologic maps. This report presents the geology and mining history of the area, together with a summary of Bureau work in progress.

Advances in mining technology have enabled deeper seams of coal to be mined from the surface. Highwall mining is frequently used to mine areas at the perimeters of a surface mine when removal of the overburden becomes uneconomical or threatens environmentally sensitive surface features. At the same time, advances in materials and technology have enabled highwall mining of coal that create blind headings over 300 meters in length. Deeper surface mining and deeper highwall penetration depths have lead to more frequent encounters with methane gas. While this coal is remotely mined from the surface, underground ignitions of methane have propagated to the surface and injured miners. Methods described in this paper show how this hazard can be correctly monitored and controlled for auger type highwall mining machines. Research indicates that small quantities of gas liberated during mining migrate to the face, creating higher gas concentrations than would otherwise be expected. Using the same principles that cause the gas to migrate to the face, researchers have shown that inert gas introduced at the collar of the hole can inert the entire hole, thus ensuring safe conditions for mining.

In the Bureau of Mines investigations of respiratory apparatus, a laboratory study of dust respirators has been conducted, and the results will be published in detail as a technical paper. Many industrial dust respirators, and many fabrics and filtering materials were tested. The principal results of these tests, and the conclusions drawn from them, are presented below. Reference to the patent literature discloses numerous types of dust respirators, mostly based on the filter principle. The present study included only representative filter-type respirators, purchased on the market.

A complete list of permissible mining equipment, rescue apparatus and gas masks tested prior to January 1, 1925, was published in Bureau of Mines Technical Paper 3761. The present list includes all equipment tested and approved by the Bureau, up to and including January 1, 1926, and supersedes the list issued as Serial 27202 in November, 1925. It covers electric air compressors, coal drills, mining machines, mine pumps, room hoists, switches, electric cap lamps, flame safety lamps, electric hand and trip lamps, flash lamp, methane indicators and detectors, blasting units, storage battery loco- motives, power trucks, self-contained oxygen breathing apparatus and gas masks. The system under which these devices were tested permits the manu- facturer, after his equipment has passed certain tests prescribed by the bureau, to mark his equipment with a seal showing that it has been "approv- ed" by the Bureau of Mines. The tests are designed to insure that the equipment has the minimum requirements for safety in use. The only object of the bureau in making such tests and publishing lists of permissible equipment is to safeguard the lives of workers and to help lessen the hazards of mining.

This report presents results of mining operations and of research and development work done during stage I of the gas combustion retorting research program conducted by the Colorado School of Mines Research Foundation and six major oil companies at the Bureau of Mines Anvil Points Oil Shale Facilities. A brief description of the gas combustion process is given and the experimental program is discussed. Included in this discussion are (1) air-gas distributor development, (2) results of gas and shale distribution studies, and (3) effects of other process variable studies. The report concludes with a statistical evaluation of experimental results. Two appendixes present a chronological account of the experimental work and tabulations of data for all retorting runs conducted during stage I.

In the 39 years from 1891 to 1929 , gas and dust explosions in the coal mines of Tennessee caused 413 deaths , or one- third of the fatalities in the mines for that period . Inasmuch as explosions caused but about 15 per cent of mine fatalities for the whole United States for this same period it is evident that the Tennessee coal mining record is by no means good , as far as explosion fatalities are concerned . Since 1926 , Tennessee has had a clear record as to fatalities from explosions ; this fact may be credited very largely to the improved conditions and safer practices in Tennessee's coal mines which have been brought about by the work of the State Inspection Department and of the Southern Appalachian Coal Operators Association directed toward adoption of safety precautions and practices most of which are advocated by the United States Bureau of Mines . Chief among these improvements are the replacement of black powder and dynamite by permissible explosives , improved ventilation , and the installation of rigid methods of inspection for gas . With the present tendency for operations to pass into the hands of responsible and informed management , and with unrelaxed care and inspection , the good record of the past several years should be maintained .

"Importance pf the small stone problem.Profitable disposal of limestone too small for making lime in the shaft kiln, is one of the most urgent problems in the lime industry. Reports received by the Bureau of Mines from 31 quarries show that the proportion of small stone varies from 2 to 40 per cent, averaging 18 per cent of gross pro¬duction, Modern methods of heavy blasting, and the use of crushers in large quarries tend to increase the proportion of small stone above that produced in early days when light blasting and hand sledging were almost universally used. There is also a growing movement toward underground operation of lime deposits, a method which in general produces a greater proportion of fines than is encountered in open-pit work. Thus the small stone problem is of growing importance, and demands the attention of every progressive lime producer. As the problem is of almost universal interest, it is urgently desired that every operator who reads this report will contribute from his knowledge or experience, answers to the questions on the final sheet.There are several markets in which small stone may be sold but in these markets the competition is generally keen and the prices low. The lime producer's chief product is lime, he understands its manufacture and marketing, and brings him a greater return than can be obtained from most limestone by-products. For these reasons it is evident that the conversion of small into lime is the nest desirable means of disposal."

"Using. ESR spin trapping methodology, evidence was obtained that fresh quartz dust in a cell-free aqueous medium generated hydroxyl - and possibly superoxide radicals. The hydroxyl radical generation potential decreases on storing of fresh quartz dust in ambient air and on the addition of either catalase, superoxide dismutase, desferoxamine, or DMSO. The role of quartz related radicals in the biochemical mechanism of acute silicosis is suggested from these findings.IntroductionThis report summarizes our electron spin resonance (ESR) detection of oxygenated radical formation by freshly crushed quartz particles in a cell-free aqueous medium, and its inihibition by radical scavengers. This study was undertaken with to elucidate the primary biochemical mechanism involved in the development of silicosis (Farber, 1983; Reiser and Last, 1979; Silicosis and Silicate Disease Committee, 1988). In particular the mechanism by which the quartz particles exert their toxic action on cells and the process(es) by which these actions progress to fibrogenesis are still not well understood (Farber. 1983; Reiser and Last, 1979;). It is generally believed that the action of quartz particles on the cell membrane is the starting point of the silicotic process (Farber, 1983; Parazzi et al., 1968). Thus the surface characteristics of the quartz particles have been the subject of several recent studies (Bolls et al.. 1983; Dalai et al., 1986; Fubini et al., 1987). Recently, We reported that mechanical crushing of quartz under normal atmosphere generates free radicals which decay with time, and that these free radicals are associated with a higher cytotoxicity of fresh quartz dust as compared to aged dust from the same stock (Dalai et al., 1986; Vallyathan et al.. 1988). The formation of Si• and. SiO--type of radicals from quartz particles crushed under atmospheric conditions has also been reported by Fubini et al. (1987). In addition, Gulumian and van Wyk (1987) have reported that quartz particles react with hydrogen peroxide (H202) to generates hydroxyl radicals (•OH) and suggested that this process might contribute to quartz's pathogenicity. However, these authors did not report the effects of metal chelators, so it was not clear as to whether the detected -OH radicals simply resulted from the Fenton reaction involving exogenous metal ion contaminants. Earlier, Marasas and Harington (1960) reported that quartz particles could function as an oxidant in a number of in vitro oxidations, including the hydroxylation of proline and lysine. They postulated that silica dusts, on reacting with water, could release H2O2 which might have the potential to react with various biological constituents and thus cause tissue damage. In fact, Gabor and coworkers (Gabor and Anca, 1974; Gabor et al., 1975) suggested that the cytotoxicity of quartz particles might be associated with the generation of some 'factor or factors possessing the properties of a free radicals capable of promoting the peroxidative chain cleavage of polyunsaturated fatty acid moieties of the phospholipids in the cell membrane. More recently Weitzman and Graceffa (1984) reported that asbestos is able to catalyze the generation of • OH radicals from H2O2. Their later work indicated that lipid peroxidation may be one of the mechanism by which asbestos produces tissue injury (Weitzman and Weitberg, 1985). This is significant because • OH radicals are capable of causing peroxidation by abstracting hydrogen atoms from cell-membrane lipids and initiating lipid peroxidation in lysosomal membrane (Fong et al., 1973). In fact, the formation of oxygenated species in reactions involving quartz was suspected since Kolbanev et al. (1980) reported the detection of H2O2 in the reaction of fresh quartz particle with aqueous medium. The present work was thus undertaken to detect the anticipated formation of some oxygenated, radical species by reactions of quartz in a cell-free media and its inhibition by radical"

This report summarizes the results of laboratory testing of copper-bearing magnetite ore from the Poorman, Bush & Brown, and Copper Center deposits, Kasaan Peninsula, Prince of Wales Island, southeastern Alaska. The ores contain minor amounts of chalcopyrite and pyrite. Intimate association of these sulfides with magnetite necessitates fine grinding for adequate liberation and rejection of copper and sulfur and for the production of high-grade iron concentrate low in objectionable impurities. Laboratory tests indicated that the optimum results can be obtained by flotation of ore ground to minus-100-mesh, with subsequent wet magnetic separation treatment of the flotation tailing. A summary of results obtained by this treatment of the three samples tested is shown in table 1.

The theory of electromagnetic fields indicates such fields could be used to detect and locate trapped miners. To be useful, the hardware of the system must meet a number of requirements, including small size, intrinsic safety, and rugged construction. Such hardware has been built, and the system has been tested by the Bureau of Mines and its contractors. These tests indicate that the electromagnetic method provides a practical means to locate miners in emergencies.

To provide coal operators with an easy-to-use stress control design procedure to minimize coal mine bumps during room-and-pillar retreat mining conducted with available continuous mining machine technology. Approach The goal of this U.S. Bureau of Mines (USBM) re¬search was to facilitate safe and efficient room-and-pillar retreat mining of bump-prone coalbeds by limiting coalbed stress adjacent to expanding gob areas through variation of section layouts. The first step was design of total-extraction pillars and their associated pillar-extraction plans. The second step was development of a coalbed strength model. The third step was development of stress limit design criteria. The fourth and final step was development of a methodology to estimate the magnitude and distribution of development and abutment loads apllied to the pillar line. The linear geometry of unlimited-width sections allowed for the accomplishment of step four with the tributary-area and linear shear-angle concepts. Increasing overburden depths required limited-width sections separated by barrier pillars to control stress within total-extraction panels. The accomplishment of step four with limited-width section designs required prediction of abutment load distributions for rectangular gobs and apportionment of these loads over mixed pillar sizes. A parametric study of the complicated interaction of various combinations of overburden depths, total-extraction panel widths, and abutment pillar widths was conducted with the USBM-developed boundary-element program MULSIM/NL.

The Bureau of Mines has developed a process to utilize waste heat from electric arc steelmaking furnace gases to preheat the ferrous material charged to the furnace. Four heat exchangers were designed, tested, and operated successfully on 1-ton electric arc furnaces. Briquets and pellets were charged continuously, both countercurrent to the hot furnace offgasses and at ambient temperatures. A comparison of the electrical energy consumed in each situation showed that preheating can lower energy requirements as much as 15.6 percent.

The Bureau of Mines has undertaken research to elucidate the chemistry involved in the efficient recovery and control of minor elements in base metal smelting operations. These elements should be effectively recovered not only because they have commercial value, but also because of their potential hazard to workers' health and the environment, if they are not controlled during processing. Cadmium, a minor metal in zinc ores, is the subject of this report. The roasting step of the complex zinc-smelting process was chosen for investigation because of the high potential for removing cadmium by volatilization during this primary stage. Commercial roaster feed materials were used to determine the sequence of reactions and the effect of these reactions on the volatilization of cadmium. Electron microprobe examination of both the commercial roaster product and oxidized sulfide solid solutions prepared in the laboratory showed that oxidized iron was of prime importance in preventing cadmium volatilization. The mechanism by which iron prevents cadmium volatilization is proposed, and a preheating technique is recommended for more effectively recovering cadmium prior to roasting.

This is the second report to be presented on th application of stress analysis to underground Mining problems. In part I2/ of this investigation the stress distribution around single openings in unidirectional and hydrostatic stress fields was analyzed. Relationships between maximum stress concentration and size and shape of openings were determined by the photoelastic method. In most cases the results were verified by mathematical methods This report considers the stress distribution in pillars formed by two or more openings in a. unidirectional stress field. Relationships between stress concentrations and size and shape of pillars are developed. Applied to mining, results snow how estimates may be made of the stress distribution in pillars, what factors cause high local stresses, and how local stress concentrations may be reduced by proper design of mine openings. The photoelastic method was used, as in Part I, to study the stress distributions, and, whenever possible, the results were verified mathematically. A description of the photoelastic method and equipment is .given in Part I.
