Based on extensive investigations on the effect of design variables on process performance, during 1982-85, Nordberg developed a 1000 hp cone crusher, with the objective of boosting crushing productivity in existing plants. The 2390 fits on the same foundation as the 7' SHD Symons cone, but is designed to give two times the capacity of the 7'. The economies of scale involved give the customer substantial cost savings. In this paper process performance of the Nordberg 2390 relative to the 7' SHD Symons cone, while crushing the same ore, will be presented. Also, implications of such a crusher scale-up to associated cost savings will be addressed.

The Hardgrove grindability index (HGI) is a widely used coal-quality parameter with applications in coal mining, beneficiation, and utilization. An understanding of the petrology of the coal is fundamental to understanding the validity of the HGI in the given application. The HGI varies with coal rank, increasing (greater ease of grinding) with increasing rank through the high-volatile bituminous rank range; and the HGI varies with changes in the maceral and microlithotype composition, decreasing with an increase in liptinite and liptinite-rich microlithotypes. In this review, the .fundamental relationship between HGI and coal petrology, and consequently its relationship to coal utilization, is examined.

In 1994 Western Mine Engineering, Inc., (WME), which is now InfoMine USA, Inc., set out to build a suite of stand-alone software programs that would provide their clients the means to estimate the costs of developing and operating mineral exploitation projects. After the first program (Sherpa for Surface Mines) was completed and launched, the developer (me) left WME to form Aventurine Engineering, Inc., in order to complete the series of applications. In the years since, four programs have either been developed or upgraded and enhanced. Applications were produced that estimate the costs of underground mining, placer mining, and project site reclamation. In addition, an application specifically designed to evaluate the economics of these projects, which was based upon a program previously developed for WME by Bill Willoughby of Willoughby & Associates, was redesigned and the code updated to a MicroSoft compatible platform. All these programs follow an engineering-based approach to cost estimating. Instead of relying on rules-of-thumb and statistical averages, they calculate all of the engineering and operational parameters needed to estimate costs. The premise is that the uncertainty associated with cost estimates should lie with the data available for the resource, not the approaches used to design and estimate the associated design parameters. In other words, it is acceptable that items such as the resource grades, deposit dimensions, ore and wall rock characteristics, and mill recovery rates carry with them some uncertainty, but parameters such as loader bucket capacities, truck speeds, pump horsepowers, hoist velocities, and grinding mill diameters can be calculated (although uncertain data might be used in those calculations). No additional uncertainties should be introduced because the cost-estimating procedure relied upon statistical manipulation or anecdotal inaccuracies. To that end, the programs rely upon calculation-based approaches to selecting and sizing machinery, assigning the required workforce, and determining the extent of the consumables needed to recover mineralized material at a rate defined by the client. And while calculation-based software is not unusual, more modern approaches work toward the development, accumulation, and manipulation of data. INITIAL CONDITIONS AND PROJECT GOALS In 2016 we had five separate programs completely disconnected from one another. Each was written in VisualBasic, a language that was falling from favor. In essence, the status in 2016 was a series of stand-alone programs (Figure 1) in which results from one needed to be manually entered as input into another.

INTRODUCTION Geotechnical models, particularly those based on the finite element method, have been available to aid in en¬gineering design of underground mining excavations for over ten years. Despite this fact there are remarkably few cases of their use in mine design documented in the literature. It is therefore to be anticipated that many potential users of these models are relatively unaware of their capabilities and limitations and also of the form and detail of geotechnical data needed for their success¬ful application. This subsection attempts to address this problem by discussing the different types of numerical models now available and by noting how some of them have been used to study a variety of problems associated with underground mining. The subsection concludes by discussing the applica¬tion of the displacement discontinuity method to the de¬sign of possible mining systems for the copper-nickel deposits of northeastern Minnesota. The object of the analyses, which were nonsite specific, was to determine the significance of geotechnical parameters, such as ini¬tial stress and rock structure, on the stability of under¬ground excavations and hence to provide guidance for future geotechnical investigation. NUMERICAL MODELS In the geomechanical design of underground mining openings, use is made of numerical models that repre¬sent or simulate the large-scale mechanical behavior of rock. There has been less interest in analysis involving fluid flow and heat transfer, but with increasing interest in such areas as in-situ retorting and solution mining it is likely that there will be a growing need for numerical models embracing and coupling all three physical proc¬esses. However, the emphasis in this subsection will be on mechanical behavior. Models which simulate such behavior will be divided into two groups: continuum models and discontinuum models. These will be dis¬cussed in turn in order that some insight into alternative solution strategies and their merits may be gained. Continuum Models Almost all geomechanical numerical models must be classed as continuum models even though particular computer codes incorporate special provisions for rep¬resenting discontinuities such as faults, bedding planes, or joints. They are continuum models because they pro¬vide solutions for cases where material behavior is governed by the differential equations of continuum mechanics. Two basic solution strategies for such equa¬tions may be identified immediately: the differential ap¬proach and the integral approach. In the differential approach a means of approximating the differential equations over the entire region of interest is sought. In the integral approach use is made of fundamental solutions from continuum mechanics, and these are used to construct a solution to the whole problem, making approximations only on the boundaries of the region of interest. The several differential and integral methods are identified in Table 1. Differential Methods: Problems in continuum me¬chanics involve the solution of three types of partial differential equations. Two of these govern the behavior in so-called initial value problems, in which variables change both in time and in space. Examples of such problems include nonsteady heat transfer and fluid flow, and stress wave propagation. The last type of partial differential equation governs the behavior in boundary value problems. In these, variation is in space but not in time. Solution of initial value problems may be achieved in two significantly different methods: implicit and ex¬plicit. The differences between these two methods will be illustrated by considering a very simple initial value problem, that of one-dimensional heat diffusion. The equation governing this process may be written as: [ ] where T is temperature, K is thermal conductivity, p is density, c is specific heat, t is time, and x is the spacial coordinate. In finite difference form this equation might be written as: [ ] where the superscripts refer to the time and the sub¬scripts to the spacial location. Several solution strategies for this equation have been used. Two important ones may be illustrated very simply by discussing the signifi¬cance of the superscript * on the right-hand side of the equation. If i + 1 is substituted for * then the second derivative of the temperature with respect to distance is evaluated at the end of the next time step (using tem¬peratures not already known). Such an approach leads to a set of equations involving unknown temperatures and a solution procedure which is referred to as being implicit. The important characteristic of the implicit procedure is that it leads to a set of equations that must be solved for each time at which the temperature dis¬tribution is required. If instead of substituting (i + 1) for the superscript *, i is substituted, the following equation is obtained: [ ] {In this case the new temperature is defined in terms of an already known temperature distribution. The data of Dunn and Martin (1978) is reanalyzed using a rigorous application of Hertzian theory to the impact process. The Young's modulus of steel required to fit their data to the theory is a factor of about 10 3, too low compared to the values quoted for steels. Thus, doubt is thrown on the validity of their experimental technique, and it is possible that their values for maximum impact force and stress are far too low.

Reduction bleaching is used to improve the brightness and color of the kaolin pigments sold to the paper and speciality industries. The bleach re¬duces the iron oxide associated with kaolin deposits to a colorless form by the following reaction: Na2S2O42+ 6Fe+3 + 4 H2O -> 2 NaHSO4 + 6Fe+2 + 6H+ (Conley et al., 1964). At present, sodium hydrosulfite is used by the kaolin industry as the reductive bleach. It is often purchased commercially as a liquid solution and added into an acidified slurry. Before the use of commercial sodium hydrosulfite, kaolin plants manufactured zinc hydrosulfite on site by adding zinc dust to sulfurous acid made by passing SO2 gas into water. This process was abandoned in the 1970s due to environmental concerns about zinc in waste waters. In the 1970s, Engelhard patented a process that made sodium hydrosulfate from an iron hydrosulfate (Tiethof, 1978). Difficulties with separating the iron hydroxide byproduct from the bleach and a decrease in the cost of commercial bleach caused the project to be abandoned.

Ion flotation of gold is a process continuing to be developed to selectively beneficiate dilute aurocyanide liquors using a novel surface­active complexing agent. Aeration of these liquors causes accumulation of the precious metal species in the froth concentrate produced in a flotation column. Successful development of ion flotation for concentrating solids-free liquors would offer a low--cost method for recovering specific metals from heap leach runoff or waste tailings solutions. The cost of constructing and operating a 100 tph gold ion flotation facility has been compared with the relevant carbon adsorption plant costs. Reductions in operating costs of 25% and installed capital of 40% are possible in relevant expenditure areas. For an 'average' processing plant, this equates to annual operating savings of A$ 170 000 and a capital saving of A$ 515 000, with a three-point increase in total (mine and plant) site DCF ROI. High gold inventories are eliminated and the number of processing steps is reduced. The application of gold ion flotation will initially be confined to heap leach operations in the western U.S.A., although use of the process in mine abandonment programmes elsewhere is also an area of future development.

INTRODUCTION Tunnel construction has always been an extremely high risk form of contracting primarily due to the inaccessability of information concerning the ground conditions to be encountered during excavation. We certainly make every effort to overcome this problem through borings, geological research, site visits, etc.; but at best owners, engineers and contractors alike are only able to interpolate a relatively small amount of factual data from isolated sections of the project into a forecast of ground conditions for the entire project. In essence this becomes an "educated guessing game" with risk to reward ratios (and subsequently the bid price) in direct relationship to the contractor's confidence in the quality of the guess. In many instances it is economically or physically infeasible to obtain information and of course the total picture is never known until the project is complete. This problem effects the contract format as well as price. The contractor's ability to handle unforseen problems on the project become extremely important as to the project's overall success. The following pages contain an actual case history of a project which affords an excellent example of the entire spectrum of situations resulting from tunnel construction in difficult ground conditions.

"Federal regulations require refuge alternatives (RAs) in underground coal mines to sustain life for 96 hours while maintaining an apparent temperature (AT) below 35°C (95°F). NIOSH research has shown that heat and humidity buildup is a major concern with RAs because they have a limited ability to dissipate heat, and high internal air temperature and relative humidity (RH) may expose occupants to heat stress. The heat transfer process within and surrounding an RA is very complex and is not easily defined analytically nor experimentally. To investigate heat and humidity buildup in RAs, NIOSH conducted multiple in-mine 96-hour tests on a 6-person metal-type RA, a 23-person tent-type RA, and a 10-person tent-type RA. The test results show that when moisture was introduced into the tests to represent perspiration and respiration from miners (wet tests), the average measured internal air temperature at midheight increased by 7.7°C, 9.4°C, and 10.5°C, and the RH approached 95%, 94%, and 88%, respectively. For the dry tests when no moisture was introduced, the average internal temperature increased by 8.4°C, 10.3°C, and 12.6°C, respectively. INTRODUCTION If an accident occurs in an underground coal mine, miners who cannot escape from the mine can enter an RA for protection from adverse conditions, such as high carbon monoxide levels. One of the main concerns with the use of mobile RAs is the thermal environment inside the RA. The metabolic heat of the occupants and the heat released by the CO2 scrubbing system will cause the interior air temperature to increase. Moreover, the humidity within the RA will increase through occupants’ respiration and perspiration and from the chemical reaction within the CO2 scrubbing system. The internal thermal conditions can subject miners to heat stress, which can lead to heat exhaustion, heat stroke, or even death depending on duration and magnitude of exposure. MSHA regulations require that RAs must be designed to ensure that the internal apparent temperature does not exceed 35°C (95°F) when the RA is fully occupied [1]. Apparent temperature (AT) is a temperature-humidity metric for the perceived temperature caused by the combined effects of air temperature, relative humidity (RH), and wind speed. It is used to assess the perception of indoor temperatures when workplaces are not sufficiently heated, cooled, or insulated to provide comfortable or healthy conditions. Before a mobile RA can be deployed at an underground mine for emergency usage, it must be tested to show that its internal AT when fully occupied will not exceed the 35°C (95°F) limit. For practical reasons, RA manufacturers usually conduct their tests at above-ground test facilities. To investigate RA thermal response in an in-mine environment, NIOSH conducted multiple 96-hour tests on a 6-person metal-type RA, a 23-person tent-type RA, and a 10-person tent-type RA in its underground coal mine facility. The work described in this paper could be used by RA manufacturers to determine a final temperature rise based solely on dry and time-reduced testing."

In the previous work (1,2), initial developments in a novel tracing technique employing specifically prepared trace particiles were fully described. The synthetic tracers produced proved to represent coal particles in specific gravity based coal washing equipment in this work, a detection system has been developed that can detect the artificial tracers in the product or refuse part of the density-separating coal cleaning units and thus indicate the performance characteristics of the circuit. This system could not only be a simple, accurate, and inexpensive replacement for the usual time-consuming washability analysis, but it could also be used for diagnostic purposes in coal separatory devices.

The Bagdad mill, built in 1977, was de-signed for a mill capacity of 36,300 tpd. An ABC (Autogenous Mill-Ball Mill-Crusher) grinding circuit was chosen. The original mill contained three completely independent grinding circuits, each containing one 9.75 m diameter by 3.96 m long fixed speed autogenous mill, one 4.72 m diameter by 6.71 m long ball mill and one 2.13 m diameter short-head cone crusher. Total installed power per grinding circuit was 8,464 kw with 5,966 kw in the autogenous mill, 2,237 kw in the ball mill and 261 kw in the crusher. (See FIGURE I--Major Equipment Flow Diagram.) The autogenous mills were designed to be able to operate semi-autogenously but were never converted to SAG. As a result of this design, only about 4,100 kw of the designed power is utilized in the autogenous mills. The grinding circuits are closed with 2.44 m by 6.10 m double deck vibrating screens. Screen oversize is returned, by a series of conveyors, underneath the mill coarse ore stockpile to feed the crushers. Variable speed belts feed the crushers and control power draw. Crusher product reports directly to the tail end of the autogenous mill feed belt. (See FIGURE 2--Typical Crusher Feed and Product Size Distributions.) Tramp iron in the autogenous mill-crusher circuit is handled in a unique way. A magnetic sensor detects the metal. This closes a gate which directs the crusher feed onto a separate belt which bypasses the crusher and returns the material to the autogenous mill feed belt.

The design of an underground mining operation requires the integration of transportation, ventilation, ground control, and mining methods to form a system which provides the highest possible degree of safety for mine personnel; the lowest cost per ton of product; the most suitable quality of final product, considering its intended use; the maximum percent recovery of the reserve, consistent with cost and subsidence rights; and the minimum disturbance to the environment. The selection of mining method is generally the controlling factor in overall mine design. Some important considerations in the choice of mining method are : 1) Depth of cover over the coal seam. 2) Strength and thickness of rock strata in the overburden. 3) Characteristics of the coal seam, including thickness, nature and location of impurities or irregularities, dip (or pitch), nature and direction of cleavage, hardness, and structural strength of the coal. 4) Nature and strength of the immediate roof (top) over the coal seam. 5) Nature and strength of the floor (bottom) under the coal seam. 6) Presence and amount of explosive gas (methane). 7) Quantity of water likely to be encountered in the mining operation. 8) Any previous or concurrent mining in seams above or below the coal seam to be worked. 9) Any restrictions on recovery required for subsidence control. Topographical and geological reports and maps of the area are usually available from state and federal geological surveys, and contain excellent general information to guide more detailed investigations. Exploratory drilling, and the plotting and analysis of the information thus obtained, are essential steps in any development program, and aid in the correlation and evaluation of information gained by other means. Other good sources of information are operating coal mines that may be in the same seam in nearby properties. Such mines may indicate in advance much about roof control, drainage, ventilation, and other mining problems.

No1ihwest Alloys produces magnesium from dolomite by reduction with ferrosilicon and aluminum. This process produces alkaline by-products in quantities that allow their consideration for use as commercial products. Two such products are dicalci­um silicate, termed as Di-Cal, and magnesium oxide, termed simply MgO. The alkaline properties of these materials offer acid neutralizing power, comparable to traditionally used lime. Thus, the materials warrant study for their use in various appli­cations where neutralization of acidic waters or process solutions are required. One such application is the treatment of acid mine waters with the objective of removing heavy metals. The chem­ical stabilities of the produced metal precipitates are critically important, from an environmental point of view, for their safe disposal in tailing ponds. The EPA regulates waste material dis­posal according to the results from TCLP testing. In this ongoing study, three mine waters were selected for treatment with Di-Cal and MgO. Lime was used as the control for these experiments. The waters were collected from the Bunker Hill mine, Berkeley Pit, and the Blackbird Mine. In addi­tion to testing the chemical stability of produced metal precipi­tates by the TCLP procedure, two other parameters were studied as well. They are: 1. the effect of water pH on stability, and 2. the effect of pH on the stability of resulting precipitates. In the first three months of study, it was found that neutral­ization with Di-Cal results in precipitates that range from equiv­alent to superior in performance as compared to those produced using lime for the waters from all three mines. An additional beneficial characteristic of precipitation with Di-Cal is the fast filtering characteristics of produced precipitates, when compared to the precipitates produced by lime. This property may be of significance when considering the improvement in drainage of tailing ponds and the volume of solids produced. This paper will discuss in detail the stability of metal precipitates with respect to various pe1iinent metals found in mine waters of Berkeley Pit (Cu, Zn, Mn, etc.), Bunker Hill (Zn, Mn, Ni and Co), and Blackbird Mine (Cu, Co).

INTRODUCTION This chapter will discuss, both from fundamental and practical points of view, the flotation of non-sulfide min- eral s. The flotation characteristics of silicates, salt-type minerals, oxides and coal will be reviewed. Dup- lication of the thorough and excellent presentations by many authors is not intended and a comprehensive list of references is included. GENERAL PRINCIPLES Collectors for Non-Sulfide Minerals : All minerals, with only a few exceptions are readily wetted by water, that is, they are said to be hydrophilic. The purpose of a collector is to coat the mineral surface and render it hydrophobic, preferring an environment of air over an environment of water. Relative to sulphide minerals, the non- sulfides are more naturally hydrophilic and therefore, require a collector with a greater chain length. This is accomplished by using a collector which has a proportionally longer hydrocarbon chain. Both anionic and cationic flotation collectors are used in non- sulfide flotation. Anionic Collectors. These are collectors which ionize in solution to give a negatively charged radical. They have the general structure R-A, where R is a hydrocarbon having 8 or more carbon atoms and A is a hydrogen or an alkali metal. The most common collectors in this group are fatty acids which could have saturated or unsaturated hydrocarbon chains. Such chains may be straight or branched. The configuration of the chains affects their solubility in water and their adsorption characteristics. In general, as the chain length increases the adsorption and the collecting power increases. However, it should be noted that the solubility decreases. The solubility of unsaturated chains is greater than that of saturated fatty acids because double or triple bonds are polar and thus attract water molecules. Solubility data for collector metal salts, even though important for a proper understanding of flotation are not available for all collector systems. Solubilities and structures of different fatty acids are reported by Fuerstenau (1982). Values for the solubility products of fatty acid salts selected from a paper by Du Rietz (1958) are given in Table 1.

Iron ore pellets use bentonite clay as binder at dosages from 0.5to 1.0% (10 to 20 pounds per ton) of moist iron ore concentrate. Bentonite is typically shipped hundreds of miles from Wyoming to the Lake Superior iron ore district. It is therefore best to know how effective a bentonite will be before it is shipped and used to bind concentrate into pellets. Current tests of bentonite effectiveness include: plate water absorption, methylene blue uptake, free swell, exchangeable cations, and glycolated layer expansion. However, these tests have many disadvantages: First, they only measure the ability of a bentonite to expand, disperse, or absorb, not how well it will bond. Second, most of the tests require up to 20 hours to perform. Bentonite only has a few minutes to interact within the concentrate before it is indurated (sintered). These long-term tests may not reflect the true behavior of bentonite. Third, the tests are tedious and often results are inconsistent between different iron ore concentrates, labs, and technicians. Fourth, some of the testing equipment is difficult to procure. Fifth, procedures are often difficult to standardize. In this paper, the Binder Effectiveness Strength (BESt) test to evaluate binders for iron ore pellets is presented that: measures the ability of bentonite to bond, requires only about two hours to complete, is simple and reproducible, and utilizes readily available equipment that has been standardized for decades.

Biooxidation processes for treating arsenical and other types of refractory gold ores are touted by many as being simple, inexpensive and environmentally friendly. One of the advantages claimed by proponents of biooxidation is that arsenic is disposed of as insoluble ferric arsenate which is suitable for long term disposal in tailings ponds. However, there is considerable controversy as to the stability of ferric arsenate with respect to re-dissolution of arsenic. In particular, there is concern that the amorphous ferric arsenate produced at low temperature is not stable, especially at the alkaline pH's required for cyanidation. There is a general lack of published information on the stability of arsenical bioleach tailings. Past work on ferric arsenate stability has usually involved purely chemical systems and the use of synthetically prepared precipitates. Upon completing the Salmita bioleach pilot plant campaign (Hackl and Wright, 1989) the author had the opportunity to test the stability of the arsenical bioleach/cyanide tailings which were produced. This paper summarizes the results of that work.

Introduction As the world economies continue to face an increased demand for natural resources to support growth in the established and developing world, the western mining industry has expanded its operations and funding of mineral developments into new and relatively unexplored regions of the world. Mining companies worldwide, in order to grow in a global economy, are now actively exploring and developing mineral resources in the emerging markets of South and Central America and most recently have expanded activities throughout Asia. South East Asia Resources (S.E.A. Resources) has recognized the vast resource potential offered by countries such as China, India and Indonesia, Asia's main driving forces of economic growth and has made a commitment to be actively involved in the area. These countries rich in mineral resources, represent some of the highly prospective and underexplored regions on earth. Their vast potential has been underestimated because of political reasons and overlooked as Central and South America still continue to be the focus of exploration for many mining companies. Most of these Asian countries have faced a prolonged period of political unrest and it has not been until recently that they have begun to enjoy a relatively stable political climate which has encouraged foreign investment, growth and economic development. These countries are in need of capital to develop their infrastructure and market economies. Their wealth in resources is one of the best methods to achieve their goals. Gold is usually one of the first targets to be extracted from developing economies. Equivalent-sized gold projects tend to cost less to develop and require less supporting infrastructure than base metals. These projects are easier to finance and, most important of all, tend to give a quicker return on investment than that offered by base metals. They also provide the economy with precious foreign exchange, international investment and the technology required to build the country. Following is a graph representing the global re-allocation of the total capital investment on the twenty largest gold projects now under construction with the total for those under construction in 1990.

5.1 INTRODUCTION Heap leach cyanidation techniques possess considerable potential for exploitation of low-grade ores, small ore bodies, mine strip waste and tailing materials where fine grinding is not necessary for good extraction. For heap leaching to be successfu1, ore must exhibit certain characteristics, as previously discussed in Chapter 1. Primary among the necessary ore characteristics is that the ore must have good permeability after being crushed or treated and stacked into heaps if uniform distribution of the cyanide leach solution is to be achieved. Gold and silver ores containing excessive amounts of clays or fines generated by crushing are some of the most difficult to treat successfully by heap leaching. The presence of excessive amounts of slimes (generally classified as minus 50-micron or 270 Tyler mesh sieve size particles) in the heap leach feed will slow the percolation flow of the leach solution, cause channeling, or produce dormant or unleached areas within the heap. This may result in unreasonably long leaching periods and poor extractions. In extreme cases, the clays or slimes can completely seal the ore heap, causing the leach solution to run off the sides of the heap rather than to penetrate the ore heap. The problem of heap leaching ores containing fines can be aggravated during stacking and preparation of the ore heaps, because natural sorting of coarse and fine material occurs during these steps (Johanson, 1978). This phenomenon results in a concentration of ore fines at the center of individual ore piles and a concomitant concentration of larger rock fragments on the lower slopes and base of the piles. When the individual piles within the heap are leveled off prior to installation of the sprinkling system which delivers the leach solution, additional segregation occurs as the fines sift through the coarser ore particles. The segregation results in localized areas or zones with marked differences in permeability. As a consequence, the leach solutions follow the path of least resistance, percolating downward through the coarse ore regions and bypassing or barely wetting areas that contain large amounts of fines or slimes. Effective utilization of marginal gold and silver resources through heap leach processing requires the development of new methods in order to achieve more uniform size distribution during ore heap preparation and better slime control during leaching.

THE FUTUREGEN CLEAN Coal project will be the recipient of $1 billion worth of federal stimulus money by way of the U.S. Department of Energy (DOE), which announced the commitment to the project. The money is expected to kick off years of further work that could finally see the project completed. One of the companies involved, Ameren Corp., said the $1.2 billion project ? which will include some private funding ? still has a number of hurdles to clear.

Since it has been demonstrated that silica caused phagocytic cells to activate lipoxygenase metabolism of unsaturated fatty acids we thought selective inhibition of this metabolic pathway might be helpful in treatment of silicosis. It is known that lipoxygenase metabolism plays a role in the pathogenesis of a variety of inflammatory and hypersensitivity conditions. Further, it is believed that the alveolar phagocytic cells which engulf silica begin the inflammatory cycle which produces the metabolic products, cytokines and growth factors which causes fibrosis in the lung. There have been indications that drugs which inhibit lipoxygenase can inhibit fibrosis from other causes. Previously, we developed a method which detects the activity of lipoxygenase metabolism by measuring free radical oxidation via light production or chemiluminescence (CL). We decided to apply this method to phagocytic cells that would be found in the lung in the presence of silica with and without newly developed inhibitors of the lipoxygenase reaction. We used drugs which inhibit lipoxygenase directly including AA861 (TAKEDA), A63162 (Abbott), and an inhibitor that scavenges free radicals produced from the lipoxygenase reaction, nordihydroguairetic acid. In addition we used an indirect inhibitor, tetrandrine. We found drug inhibition of silica activated CL from alveolar macrophages, neutrophils and human leukemia (HL-60) cells differentiated with DMSO. Nordihydroguairetic acid 2x10-5M produced almost complete inhibition in all three cell types. In human leukemia cells (HL-60) A63162 and tetrandrine were equally inhibitory at 42% while in human neutrophils A63162 and tetrandrine were less active 22 and 10% respectively. In macrophages A63162 and AA861 were less than 50% inhibitory to cellular CL at 2xl0-5M. Direct assay of tetrandrine (2x10-5M) with sodium arachidonate and soybean lipoxygenase reveals no inhibition. All antilipoxygenase drugs regardless of mechanism were inhibitory to cellular chemiluminescence from all three types of phagocytic cells. These drugs alone or in mixtures could prove useful in treatment of silicosis.
